摘  要:以压电传感器为检测装置,采用等离子体聚合膜沉积技术和纳米金亚单层自组装技术设计传感器的敏感界面,研制了一种新的日本血吸虫免疫传感器。先在石英晶振上沉积正丁胺等离子体聚合膜,利用膜上氨基与纳米金的共价键合作用组装一纳米金亚单层,得到可固定日本血吸虫抗原和易于传感器再生的界面,以牛血清白蛋白(BSA)封闭晶振上的非特异性吸附位点,用于直接响应感染兔血清中日本血吸虫抗体。探讨了纳米金的自组装和抗原包被等主要检测条件的影响;将传感器应用于不同感染程度兔血清样品的检测,结果令人满意。A A piezoelectric immunosensor based on a novel biomolecular immobilization strategy has been developed to determine Schistosoma - japonicum antibody (SjAb) in infected rabbit serum (IRS) . On the surface of quartz crystal microbalance (QCM), plasma polymerized films (PPF) of n - butyl amine were firstly formulated by glow - charge and then nano - Au semi-monolayer by covalent interaction between Au and amine on PPF. The resulting interfacial design which could favor the easy regeneration of the crystal, was used to immobilize Schistosoma -japonicum antigen (SjAg) to detect SjAb in IRS after being healed by BSA. The main conditions of the assembly of nano - Au and the immobilization of SjAg were investigated. Compared with the direct gold attachment procedure, the developed procedure shows improved features, including larger amount and higher immunoactivity of immobilized antibody. Subsequently, the developed immunosensing system which is sensitive to dilution ratio of SjAb as low as 1 : 5000, was evaluated on several practical IRS samples with satisfactory analytical results, indicating a promising alternative tool for clinical diagnosis of Schistosoma -japonicum disease.
