摘  要:目的探讨活血化瘀法对急性心肌梗死(AMI)大鼠骨髓干细胞(CD34阳性细胞)动员入血及定向归巢于梗死心肌的影响。方法实验用SD大鼠48只,急性心肌梗死模型造模成功32只,随机分为4组,分别为中药+G-CSF组、G-CSF组、模型组、假手术组,每组8只。各组均于造模3 h后给于对应药物,1次/d,连续6 d给药。7 d后,检测外周血CD34阳性细胞数,观察苏木素-伊红(HE)染色病理情况,免疫组化检测心肌CD34阳性细胞校正吸光度(IOD)值、及Ki-67阳性细胞数。采用SPSS 17.0软件进行统计处理,计量资料以(x±s)表示,多组间均数比较采用完全随机设计的单因素方差分析,两两比较采用LSD检验,P<0.05为差异有统计学意义。方差不齐时采用非参数检验,P<0.05为差异有统计学意义。相关性分析采用Spearman相关分析。结果药物组较模型组CD34阳性细胞校正光密度明显增高,中药+G-CSF组G-CSF组CD34阳性细胞校正光密度明显增高,中药+G-CSF组比西药组Ki-67阳性细胞数的表达高(P<0.05)。结论活血化瘀法能促进骨髓干细胞细胞动员入血,并定向归巢于梗死心肌,并能影响心肌组织细胞的新生,是一种良好的骨髓干细胞动员剂。Objective To explore the relationship of promoting blood circulation and removing blood stasis with bone marrow stem cell(CD34 positive cells) mobilizing into blood and directional homing to infarcted myocardium among rats with acute myocardial infarction(AMI).Methods 48 Sprague Dawley rats were used in the study,and 32 rats were made models successfully.32 rats were randomly divided into 4 groups evenly:traditional Chinese medicine group plus G-CSF group,G-CSF group,model group,sham operation group.Rats were given corresponding drugs after 3 hours of modeling for once per day and continuous administration of 6 days.At the 7'' day,CD34 positive cell in peripheral blood were detected,and hematoxylin eosin(HE) staining pathology were made.Immunohistochemical detection of myocardial IOD of CD34 positive cells and the number of Ki-67 positive cells were conducted.SPSS 17.0 was used to analyze the data.Continuous variables were described using mean±SD.Single ANOVA was used to compare mean difference of more than 2 groups,and LSD test was used between 2 groups.P<0.05 was considered as statistically significant.Spearman correlation was used to analyze the relationships between different variables.Results Compared with model group,IOD of CD34 positive cells among the drug group was significantly higher.Compared with G-CSF group,IOD of CD34 positive cells among traditional Chinese medicine plus G—CSF group were enhanced significantly.The expression of Ki—67 positive cells among traditional Chinese medicine plus G—CSF group is higher than among western medicine group(P>0.05).Conclusions The method of promoting blood circulation and removing blood stasis can promote mobilizing bone marrow stem cell into the blood,directional homing to infarcted myocardium,and affecting the cardiac tissues of new born cells.This is a good bone marrow stem cell mobilization agent.
