妊高征胎盘组织中胰岛素样生长因子-Ⅱ的定位及定量分析

摘　　要：目的　探讨胎盘组织中胰岛素样生长因子 -Ⅱ (IGF -Ⅱ )定位 ,评估IGF -Ⅱ在妊娠高血压综合征 (妊高征 )中的作用。方法　用免疫组化法 ,对 30例正常孕妇、2 5例妊高征孕妇的胎盘组织进行IGF -Ⅱ定位 ,并用计算机图像分析系统进行定量分析比较。结果　⑴胎盘IGF -Ⅱ主要位于绒毛小叶的合体滋养细胞及细胞滋养细胞。此外 ,也存在于羊膜绒毛层 ,但染色较以上两种细胞明显减弱。⑵与正常胎盘组比较 ,妊高征组IGF -Ⅱ的平均光密度显著降低 (t =2 .30 ,P <0 .0 5 )。结论胎盘组织中IGF -Ⅱ主要位于胎盘小叶的合体滋养细胞、细胞滋养细胞 ;胎盘组织合成和分泌IGF -Ⅱ的多少 ,与妊高征的发生有关。Objective: To investigate the immunohistochemical localization of insulin-like growth factor-Ⅱin normal human placenta and to evaluate the role of IGF-Ⅱin pregnancy induced hypertension syndrome (PIH). Methods: Control group was consisted of thirty normal pregnancies. Study group was consisted of tweenty-five women with PIH. The cellular distribution of IGF-Ⅱin human placenta was determined by immunohistochemistry. The mean density was determined by image analysis technique. Results: IGF-Ⅱwas found in syncytiotrophoblast, cytotrophoblast and amnio-chorion. The strongest immunoreactivity was observed in the syncytiotrophoblast and cytotrophoblast. The mean density of PIH group was significantly lower than that of normal group. Conclusion: These results suggested that IGF- Ⅱin human placenta was principally synthesized and secreted by syncytiotrophoblast, cytotrophoblast and cells of amnio-chorion. IGF-Ⅱmay has the key role for PIH forming.
