摘  要:目的探讨脑挫伤早期细胞凋亡相关基因Bcl-2、Fas/Fas -L的表达变化规律及神经细胞凋亡的调控机制。方法用光镜观察脑挫伤后脑组织的形态学变化 ,同时应用免疫组织化学SABC法结合图像分析检测脑挫伤早期不同时程Bcl-2、Fas/Fas -L的表达变化规律。结果伤后 8h在脑挫伤周围可见典型的凋亡神经细胞 ;伤后 3 0min损伤灶周围神经细胞出现Bcl-2、Fas/Fas-L阳性表达 ,以后呈逐渐上升趋势且表达范围亦逐渐扩大 ,其中Bcl-2蛋白表达在伤后 4h达高峰 ,随后呈下降的趋势 ,至伤后 1 0 -1 2h其表达程度与伤后 1h相比无明显差异。结论神经细胞凋亡参与脑挫伤后病理形态学改变 ,神经细胞凋亡的形成可能与促凋亡调控基因和抑凋亡调控基因的表达不平衡有关。Objective To investigate the expressive variety of apoptotic regulating gene Bcl-2 and Fas/Fas-L and the molecular mechanism of neuronal apoptosis in the earlier period of cerebral contusion.Methods The morphologic feature of brain tissue was observed under microscopy and immunohistochemistry technique and imagine analysis were applied to detect the expression of Bcl-2 and Fas/Fas-L.Results The typical apoptotic cell was seen 8 hours after cerebral contusion.The expression of apoptotic regulating gene was noted after 30 minutes and keep increase subsequently.The expression of Bcl-2 reached maximum level at 4 hours after injury and then declined.Conclusion Cerebral contusion can facilitate the occurance of neuronal apoptosis.The mechanism of neuronal apoptosis is correlative with the unbalanced expression of apoptotic regulating genes.
