摘 要:目的 研究葡萄糖对培养血管内皮细胞表达的影响。方法 大鼠肺微内皮细胞在高浓度糖(高糖)和正常浓度糖培养基中培养24 h,作基因芯片分析。结果 在超过4000个基因芯片显示的基因中,高糖引起509个基因表达增加,961基因表达减少。其中细胞基质金属蛋白酶(matrix metalloproteinase,MMP)10基因表达增加6.06倍,Wilcoxon配对符号秩检验P=0.0001。结论 MMP10在高糖环境下培养的大鼠肺内皮细胞表达增加,可能对糖尿病慢性并发症的形成过程起一定作用。Purpose: To investigate the profile of elevated glucose induced responses of endothelial cell (EC) at the gene expression level. Methods: Rat pulmonary microvascular EC were exposed to DMEM medium with normal (5 mmol/L) or High concentrations glucose (20 mmol/L) for 24 hours for oligonucleotide microarray analyses. And the results were analyzed with the Wilcoxon's signed rank test. Results: Of over 4 000 genes on the microarray, expression levels of 509 and 961 genes appeared to be upregulated and downregulated by high concentration glucose, respectively. Matrix metalloproteinase 10 (MMP 10) expression was increased by 6.06-fold (P = 0.000). Conclusions: MMP-10 expression was upregulated in Rat pulmonary EC cultured in high glucose concentration, elevated MMP 10 may contribute to diabetic complications.
