鲨鱼和魟鱼肝铁蛋白电泳纯的制备技术

摘　　要：采用硫酸铵分级盐析、DEAE阴离子交换层析、SephacrylS-300凝胶层析技术分离纯化鲨鱼肝铁蛋白 (LiverferritinofSphymazygaena,SZLF)和鱼肝铁蛋白 (LiverferritinofDasyatisakajei,DALF)。电子显微镜技术揭示了SZLF和DALF的分子结构 ,阐明了两者蛋白均由蛋白壳和铁核组成。柱层析和电泳技术拓展了电泳纯的SZLF和DALF制备方法 。Liver ferritin of Sphyma zygaena (SZLF) and Liver ferritin of Dasyatis akajei (DALF) were isolated by ammonium sulfate fractions, anion exchange chromatography with DEAE-52 column and gel exclusion chromatography with sephacryl S-300 column. Using the electron microscope, the molecular structure of both SZLF and DALF could be observed showing that both proteins consisted of a protein shell and iron core. We were able to develop the electrophoretic preparation method for SZLF and DALF using column chromatography and electrophoretic technology allowing for the preparation of other high-purity ferritins.
