摘  要:目的:探讨采用葡萄糖消耗法进行快速念珠菌药敏试验的实用性和优越性。方法:采用葡萄糖消耗法快速检测104株念珠菌对氟康唑和伊曲康唑的敏感性,并和NCCLS M27-A方案微量法进行比较。受试菌株主要分离自临床深部真菌感染的患者,包括白念珠菌(C. albicans)45株、近平滑念珠菌(C. parapsilosis)24株、热带念珠菌(C.tropicalis)17株、克柔念珠菌(C.krusei)10株、光滑念珠菌(C. glabrata)8株。结果:对于氟康唑,两种方法在±1个、±2个稀释度范围内的一致性为76.0%和89.4%;对于伊曲康唑,两种方法的一致性分别为82.7%和89.4%。结论:葡萄糖消耗法和NCCLS M27-A方案微量法的一致性较高,可以用于念珠菌药敏的快速测定。Objective: To investigate the availability and superiority with a new colorimetric assay for antifungal (Candida) susceptibility testing based on glucose utilization. Methods: The new colorimetric assay was used to determine the MICs of fluconazole and itraconazole for 104 clinical isolates and compared with M27-A methods. Most of the tested isolates were isolated from patients with deep mycoses, included C. albicans(45 isolates), C. parapsilosis(24 isolates), C. tropicalis(17 isolates), C. krusei(W isolates), and C. glabrata(8 isolates). Results: For all the isolates tested, fluconazole and itraconazole demonstrated favorable consistencies in MICs between the two methods. For fluconazole the consistencies within 1 and 2 twofold dilutions were 76.0% and 89.4% respectively. For itraconazole the consistencies within 1 and 2 twofold dilutions were 82.7% and 89.4% respectively. Conclusion: The new colorimetric assay based on glucose utilization is a reliable and rapid method which can be used for antifungal susceptibility testing of Candida spp..
