摘 要:目的 :通过研究人工合成成骨生长肽 (sOGP)对红系造血祖细胞的体外促增殖作用 ,初步探索sOGP促造血活性的作用环节。方法 :将正常小鼠和人骨髓有核细胞在标准红系祖细胞基础培养基中进行培养 ,设定sOGP的不同浓度梯度 ,于培养 4 8h后计数小鼠CFU -E集落数 ,培养第 4天与第 14天计数人CFU -E、BFU -E集落数 ,将数据与空白对照组及EPO阳性对照组进行比较。结果 :sOGP能够有效促进小鼠CFU -E及人CFU -E、BFU -E的体外增殖 ,量效关系明显 ,其有效作用浓度范围为 10 -16~ 10 -10 mol/L ,高峰浓度约为 10 -14 ~ 10 -12 mol/L ,呈低浓度、广范围及双相作用的特点。但是 ,sOGP对红系造血祖细胞的体外促增殖效果要弱于阳性对照药EPO。结论 :sOGP能够在体外有效地促进红系造血祖细胞的增殖 ,可能是通过对骨髓基质细胞的作用而促进造血和造骨。Objective: To evaluate the promotive property of the proliferation of precursor of erythrocyte by synthetic osteogenic growth peptide (sOGP) in vitro. Methods: Bone marrow karyote of normal mice and human were employed to culture in basic medium with standard precursor erythrocyte and different gradient concentration of sOGP. Number of mice CFU-E colonies were counted after 48 hours of culture; human CFU-E and BFU-E colony numbers were counted on the fourth and fourteenth day respectively. Data were compared with vacuity negative control group and EPO positive control group. Results: In vitro, sOGP significantly promotes the proliferation of mice CFU-E and human CFU-E and BFU-E colonies, with obvious dose-effective relationship, and the affect shows feature of low concentration, broad coverage and double phase. The effective cultural concentration range was 10 -16 ~10 -10 mol/L, with the peak concentration range as 10 -14 ~10 -12 mol/L. However, the in vitro effect of sOGP on precursor of erythrocyte was weaker than that of positive control drug of EPO. Conclusion: sOGP could promote the in vitro proliferation of precursor erythrocyte effectively, possibly, sOGP stimulates hematopoiese and osteogenesis via the way of bone marrow stroma cell.
