摘  要:目的 室管膜下层 (Subventricularzone ,SVZ)神经干细胞分化过程中的Bmp4启动子活性分析及其与神经元分化的关系。方法 在构建Bmp4启动子———红色荧光蛋白报告载体基础上转染SVZ神经干细胞 ,动态地观察报告基因在SVZ神经干细胞分化过程中的表达 ,并进行显微计数和神经元标志物流式细胞仪检测。结果 SVZ神经干细胞贴壁分化过程中具有较高的Bmp4启动子活性 ,48h阳性率达到 8.6%,且明显高于皮层神经干细胞。SVZ神经干细胞的Bmp4启动子活性与神经元分化高度正相关 (r =0 .80 5 ,P <0 .0 5 ) ,皮层神经元分化与Bmp4启动子活性相关不明显。结论 采用启动子活体荧光标记 ,能够较好地反映正常情况下内在蛋白的表达情况 ;Bmp4启动子活性与SVZ神经干细胞向神经元的分化高度相关 。Objective To analyze the activity of Bmp4 (Bone morphogenetic protein) promotor in differentiation of subventricular zone(SVZ) neural stem cells and its relation to neuron differentiation. Methods SVZ neural stem cells were transfected using reporter vector conjugating Bmp4 promotor and red fluorescence protein. Expression of red fluorescence protein was traced continuously in the living cells and the positive cell number was counted microscopically. Neuron specific enolase (NSE) positive cells were also detected by flow cytometry. Results Higher activity of Bmp4 promotor in differentiation of SVZ neural stem cells was detected, as compared with that of the cortex neural stem cells. The maximal positive ratio reached 8.6% in SVZ. Significant relation between Bmp4 promotor activity and neuron differentiation was found in SVZ neural stem cells( r =0.805, P <0.05), but not in cortex. Conclusion Fluorescence labeling of the living cells using Bmp4 promotor conjugating red fluorescence protein is a good way to reflect the integrated protein expression. Bmp4 promotor activity is closely related to neuron differentiation, suggesting that Bmp4 plays an important role in differentiation of SVZ neural stem cells.
