摘 要:目的 观察创伤后不同时相大鼠腹腔巨噬细胞分化表型与吞噬功能的动态变化。方法 采用创伤失血模型 ,运用流式细胞仪分析大鼠巨噬细胞标志性抗原ED1、分化抗原ED2的表达情况 ,并用无色孔雀绿法测定大鼠腹腔巨噬细胞吞噬功能变化。结果 创伤后大鼠pMΦ分化表型ED2表达比例降低 ,于创伤后 12h至最低 ,伤后第 2天有短暂升高 ,但严重创伤后至第 7天仍未完全恢复至正常 ,无单纯的ED2表达 ;无色孔雀绿法测定创伤后非特异吞噬功能下降 ,于创伤后第 1天达最低点 ,LPS刺激可持续减弱吞噬功能。结论 创伤影响巨噬细胞的分化状态 ,伴随表型改变而出现的分化状态不同可能是吞噬功能减低的原因之一。Objective To observe the serial changes of differentiatial phenotype and phagocytosis of rat peritoneal macrophages follo wing traumahemorrhage. Methods The expression condition of ED1,ED2 on peritoneal macrophage was assayed by flow cytometry , and phagocytosis was determined by leucomalachite green (LPS group and non LPS group, the LPS final concentration is 10 μg/mL) at various time points following traumahemorrhage.Results Expression of differentiatial phenotype ED2 of peritoneal macrophage decreased and reached to the lowest point at the 12th hour, and increased briefly on the 2nd day, but was still lower than normal level on the 7th day following traumahemorrhage.No expression of pure ED2 was found phagocytosis function decreased and reached to the lowest point at the 1st day following traumam and phagocytosis with LPS stimulation decreased persistently to the last. Conclusion Differentiatial state of macrophages is associated with trauma and the change of phenotype may be one of the factors to the decrease of phagocytosis.
