摘 要:目的 观察高晶体 高胶体渗透压混合液对人脐静脉内皮细胞 (HUVEC)细胞容积和存活率的影响。方法 取HUVEC株ECV 3 0 4,分为对照组和实验组。对照组细胞先置于缺氧( 95 %氮气 ,5 %CO2 )环境中 8h ,再在常氧状态下 ( 95 %空气 ,5 %CO2 )培养 16h。实验组细胞先置于高晶体 高胶体渗透压环境的培养液中 15min ,其他步骤同对照组。并分别于实验前、实验开始后 15、60min、8、16及 2 4h测两组细胞的容积和存活率。结果 未经高渗混合液处理的对照组细胞在缺氧和氧再灌后明显进行性肿胀 ,为正常细胞容积的 111.1%~ 14 7.8% (P <0 .0 5或 0 .0 1) ,细胞存活率进行性降低 ,分别为 86.3 %~ 72 .2 % (P <0 .0 5或 0 .0 1)。经高渗混合液处理的实验组细胞仅在 15min时有明显的缩小 (P <0 .0 5 ) ,为正常细胞容积的 76.2 % (P <0 .0 5 ) ,随后基本恢复至实验前的水平 (P >0 .0 5 ) ,其细胞存活率无明显变化 (P >0 .0 5 )。结论 高晶体 高胶体渗透压混合液可以消除内皮细胞在缺氧 /氧再灌状态下的肿胀 ,维持较高的细胞存活率。Objective To investigate the effect of hypertonic -hyperoncotic solution on cell volume and survival rate in human umbilical vein endothelial cell (HUVEC).Methods ECV-304 were selected and randomly divided into control group and experimental group.In control group,cells were cultured in hypoxia environment (95% nitrogen,5% carbon dioxide) for 8 h,and then in normal environment (95% air,5% carbon dioxide) for 16 h.In experimental group,cells were cultured in hypertonic hyperoncotic solution for 15 min firstly,then managed as control group.Cell volume and survival rate were detected just before experiment and 15 min,60 min,8 h,16 h,24 h after beginning of experiment in all groups.Results In control group,the cell volume became inflated to 111.1%~147.8% of normal cell volume and survival rate of cell was progressively decreased to 86.3%~72.2% respectively in hypoxia period and hypoxia reperfusion period (P<0.05 or P<0.01).In experimental group,cells became deflated and the cell volume was reduced to 76.2% of normal cell volume at 15 min only (P<0.05),then restored to the level just as well as before experiment (P>0.05),survival rate did not change significantly (P>0.05).Conclusion Hypertonic-hyperoncotic solution can diminish the inflation of endothelial cells in hypoxia- reperfusion period and maintain high survival rate.
