摘  要:目的 探讨雪旺氏细胞移植对电针损伤的大鼠中脑网状结构的修复作用。方法 新生大鼠坐骨神经雪旺氏细胞体外培养 ,5溴 脱氧尿嘧啶 (BrdU)标记后移植至电针损伤的大鼠中脑网状结构。免疫组织化学方法观察不同时间BrdU、生长相关蛋白 43 (GAP 43 )的表达及髓鞘染色观察新生的髓鞘。结果 雪旺氏细胞移植后 8个月仍可见BrdU阳性细胞 ,且细胞数增加 2 3 % ,并主要向损伤侧大脑皮层迁移 ;与对照组相比 ,移植后 1个月损伤的中脑神经元GAP 43的表达有显著意义 (P <0 .0 5 ) ;在损伤的脑干区域可见新生的髓鞘。结论 雪旺氏细胞对损伤的中脑网状结构有促修复作用。Objective To explore the ability of grafted Schwann cells to promote restore of injured brain stem reticular structure in rats.Methods Schwann cells originated from sciatic nerves of 1 to 2-day-old rats were expanded and labeled with BrdU in vitro,transplanted into rat brain stem reticular structure injured by electric needle stimulus.Immunohistochemistry and myelin-staining were used to detect the expression of BrdU,GAP-43 and new myelination respectively.Results BrdU positive cells could be identified for up to 8 months and the number increased by about 23%,which mainly migrated toward injured ipsilateral cortex.The GAP-43 expression reached its peak one month after transplantation and was significantly increased as compared with control group (P<0.05).New myelination could be seen in destructed brain stem area.Conclusion The transplantation of Schwann cells could promote the restore of injured brain stem reticular structure.
