摘  要:目的 观察血流切应力对植入的异种生物瓣膜组织宿主内皮细胞的生长形态和排列的影响。方法 将戊二醛和L -谷氨酸 (L -G)处理的牛心包片进行体外内皮细胞种植培养 ,并同时将同样处理的牛心包片置入猪的右心房近三尖瓣处 ,扫描电子电镜观察心包片宿主内皮细胞生长的形态学特征。结果 牛心包片体外内皮细胞种植培养及猪右心房三尖瓣处移植后 ,心包片表面内皮细胞生长良好。体内移植牛心包片内皮细胞从缝合端向游离端生长 15天的过程中 ,细胞形态和排列随着血流机械应力的影响 ,细胞从近似圆形铺路石状逐渐向长梭形变化。缝合端内皮细胞呈一致的长梭形 ,内皮细胞生长区中部细胞呈梭形、卵圆形、圆形混杂 ,内皮细胞生长端细胞呈铺路石状 ,与培养板孔中培养的心包片表面连续单层内皮细胞 (来源于人脐静脉内皮细胞 )形态一致。结论 在在体状态下 ,处于三尖瓣处的异种生物瓣膜上的内皮细胞经血流作用 ,变形为长梭形 ,排列方向趋向于血流切应力作用的方向 。Objective To investigate the influence of the hemodynamic force on the morphometric feature and arrangement of host endothelial cells over the xenograft in vivo.?Methods Bovine pericardium flap treated with 0.5% gluaraldehyde and 8% L-glutamic acid was implanted in the culture of human vascular endothelial cells harvested from umbilical vein in vitro. Meanwhile, the pericardium flap treated the same way was implanted into the pig right atrium in vivo. The morphometric features of the implanted bovine pericardium were studied under scanning electron microscopy 15 days later.?Results Morphological analysis demonstrated that the growth of endothelial cells over the bovine pericardium was good enough both in vitro and in vivo. In the pig right atrium, the form and arrangement of the endothelial cells on the bovine pericardium had been changing under the local hemodynamic forces in the 15 days. The form of endothelial cells transformed from round and oval to long fusiform: i.e. long fusiform at the suture end of the pericardium flap, mixed fusiform and oval at the middle zone of endothelial cells, and paved-stone form at the free end of the flap which was similar to what was seen in vitro.?Conclusion Under the hemodynamic forces at the spot of tricuspid valve, the host endothelial cells spread over the bovine pericardium flap become elongated and aligned in the direction of flow. The phasic changes in morphometric feature of endothelial cells in vivo were different from those under steady flow seen in vitro.
