摘  要:目的 探讨血管内皮生长因子 (VEGF)在食管癌中的表达规律及其在血管生成和肿瘤侵润转移中的作用机制。方法 应用原位杂交和免疫组织化学方法 ,检测 5 8例食管癌组织中VECF的mRNA和蛋白的表达 ,并用CD34标记癌组织血管内皮细胞 ,计数肿瘤内微血管密度 (HVD)。结果 食管癌细胞高表达VBGFm RNA和蛋白 ,阳性率分别为 6 2 .1% (36 / 5 8) ,6 0 .3% (35 / 5 8) ,VECF的表达与食管癌的浸润深度、淋巴结转移及THH分期密切相关 ,而与分化程度和组织分型无关 ;VECF阳性表达组的微血管密度明显高于低表达组 (P <0 .0 1)。结论 VEGF可促进食管癌血管形成 ,同时也促进肿瘤的浸润和转移 ,可作为反映食管癌进展的生物学指标。Objective: To investigate the expression of vascular endothelial growth factor (VEGF) in esophageal carcinoma and its effect on angiogenesis,tumor invasion and metastasis. Methods: In situ hybrization and immunohistochemistry were used to detect the mRNA and protein expression of VEGF and microvessel density (MVD) in 58 cases of esophageal carcinoma. Results: VEGFmRNA and its protein were highly expressed in tumor cells of esophageal carcinoma with the positive rates were 62.1 %(36/58) and 60.3 %(35/58) respectively VEGF expression was closely correlated with depth of invasion, lymph-node metastasis and TNM stage, but the relation to the differentiation and histclogic types was insignificant. The MVD in VEGF- positive group was considerably higher than that in VEGF-negative one (p<0.01) . Conclusions:VEGF can promote angiogenesis, invasion and metastasis of esophageal carcinoma. VEGF is an useful biological predictor of progression in esophageal carcinoma.
