摘 要:目的:探讨由营养因子碱性成纤维细胞生长因子(bFGF)、表皮生长因子(EGF)及N2添加剂所组成的配方中最适合神经干细胞存活和增殖的组合。方法:分离培养大鼠脑神经干细胞,免疫荧光细胞染色加以鉴定,采用不同组合配方的培养液培养细胞,通过计数神经干细胞球数,判定其对神经干细胞存活和增殖的影响。结果:在bFGF,EGF和N2组成的8种不同配方培养液中,bFGF组是神经干细胞存活和增殖的最适配方,是8周中与对照组比较所有P<0.05的惟一配方;EGF具有促进神经干细胞分化的作用,而促增殖能力不如bFGF;相比之下,N2对神经干细胞增殖的影响最弱。结论:bFGF对神经干细胞增殖及较长期存活具有重要的作用,其作用较EGF和N2强,EGF还具有促神经干细胞分化的作用。AIM:To explore combination,which suits the survival and proliferation of neural stem cells best,composed by the basic fibroblast growth factor(bFGF),epidermal growth factor(EGF)and N2. METHODS:Neural stem cells were isolated from rat's brain,and the features of cells were characterized by immunofluorescence staining.The effects of different culture medium containing bFGF,EGF or N2 on survival and proliferation of cells were determined by neurosphere counting. RESULTS:Among the eight different medium groups composed by bFGF,EGF and N2,the bFGF group was preferable and was the only group in which P was less than 0.05 during the 8 weeks,EGF could promote neural stem cells to differentiate but its ability of proliferation was not as good as that of bFGF,and N2 had the weakest effect on the survival and proliferation of neural stem cells. CONCLUSION:bFGF is very important to the survival and proliferation of neural stem cells,it is more effective than EGF and N2,EGF can also promote the differentiation of NSC.
