摘 要:利用形态学、细胞学、A-PAGE和RAPD方法,对5个小麦-中间偃麦草(Thinopyrumintermedium)双体异附加系Line1、Line4、Line10、Line14和Line15进行了鉴定。细胞学鉴定结果表明,它们根尖细胞染色体数目为2n=44,花粉母细胞减数分裂中期 (PMCM )染色体构型为2n=22 ,具有高度的细胞学稳定性;形态学鉴定和A-PAGE电泳分析证明,Line1和Line15可能附加了中间偃麦草第7部分同源群的染色体,Line10和Line14可能附加了中间偃麦草第1部分同源群的染色体,Line4则可能同时存在多种染色体变异;RAPD分析表明,在供试的100个随机引物中,有5个引物S21、S29、S57、S121和S152能够在亲本中间偃麦草和双体异附加系中稳定扩增出特异带型,并可作为异附加系所附加染色体的特异RAPD标记。5 wheat-Thinopyrum intermedium alien disomic addition lines (Line 1,Line 4,Line 10,Line 14 and Line 15) were identified by using the methods of morphology,cytology,A-PAGE and RAPD analysis.Cytologic results showed that the number of chromosomes in their root tip cells was 2n=44,the chromosome configuration at PMC MⅠ was 2n=22Ⅱ.Results of morphology and A-PAGE showed that Line 1 and Line 15 might contain Thinopyrum intermedium chromosomes of homoeologous group 7,the added alien chromosomes of Line 10 and Line 14 might belong to homoeologous group 1,and other chromosome aberration might also occur in Line 4.100 random primers were used for RAPD amplification,the specific bands of Thinopyrum intermedium parent could be observed in the electrophoretogram of 5 disomic addition lines by 5 of them,which could be used as RAPD markers for alien addition chromosomes.
