氯沙坦对动脉粥样硬化早期家兔血管增生及MAPK表达的影响

摘　　要：目的　探讨血管紧张素Ⅱ受体拮抗剂氯沙坦对加速性动脉粥样硬化 (AS)早期家兔血管增生及MAPK蛋白表达的影响。方法　采用组织学和免疫细胞化学方法评价加速性AS早期家兔血管组织学、PCNA及MAPK蛋白表达的改变。结果　AS组血管内膜与中膜厚度比值、内膜PCNA阳性细胞数及MAPK蛋白表达平均光密度积分值均显著增加 ,氯沙坦干预组内膜与中膜厚度比值较AS组显著下降 ,PCNA阳性细胞数明显减少 ,血管壁MAPK蛋白表达水平明显下降 (P值均 <0 .0 0 1 )。结论　氯沙坦干预可抑制血管增生时MAPK蛋白表达 ,减轻加速性AS病变血管内膜增生 。Objective To investigate effects of losartan on expression of mitogen activated protein kinases (MAPK) and vessel proliferation in a rabbit model of early accelerated atherosclerosis.Methods Expression of proliferation cell nuclear antigen(PCNA) and MAPK protein were detected by immunohistochemistry.Results The ratio of vascular intimal to medial thickness,numbers of positive PCNA cells,and mean OD values of expression of MAPK protein in AS group were singnificantly increased than in control group.There was a significant decrease in the ratio of vascular intimal to medial thickness,numbers of positive PCNA cells and mean OD values of expression of MAPK protein( P <0.001) in losartan pretreated group.Conclusion Losartan inhibits vascular MAPK expression and intimal proliferation,which may play an important role in prevention of athrosclerosis.
