摘 要:怀孕小鼠血清花环抑制滴度显著地高于未孕鼠(p<0.01),说明怀孕血清中存在早孕因子。山羊血清经DEAE—纤维素柱层析、0.05molPBS液(pH7.0)洗脱,收集最初5ml洗脱液并用蔗糖进行浓缩,然后采用改良的小鼠脾细胞花环抑制实验对其花环抑制能力进行测定,结果发现怀孕山羊血清的花环抑制滴度显著地高于未孕山革、发情山革和公革(p<0.001),说明在怀孕山羊血清中也存在早孕因子。进一步研究发现在配种后24h时,山羊血清中即可测出早孕因子活性。而且,怀孕山羊血清早孕因子活性在配种后21天内维持在较高水平,结果提示血清早孕因子可做为山羊早孕诊断的指标。Aiming at determining serum early pregnancy facteor and studing its characteristics in goats, a modified rosette inhibition test witth mouse spleen cells was set up in present study.The rosette inhibition titres of pregnant mouse sera were significantly higher than that of non-pregnant mouse sera(p<0.01), which indicated early pregnancy factor did exsist in pregnant mouse serum. After fractionated with DEAE-cellulose and eluted with pH7.0, 0.05mol PBS buffer, the mean rosette inhibition titre in caprine pregnant sera was significantly higher than that of non-pregnant, oestrus, and male goats (p<0.001), but serum rosette inhibition titres among the goats under late three phsiologicai status were observed at the same level. These results showed that early pregnancy factor also exsisted in pregnant serum in goats. Moreover, the early pregnancy factor activity appeared within 24 h after mating, and sustained at pregnant level untill day 21 after mating. These results implied that detection of caprine early pregnancy factor in serum could be used in early diagnosis of pregnancy in goats.
