摘  要:目的 :以60 Co γ 射线照射大鼠损伤为模型 ,研究硫辛酸对大鼠谷胱甘肽 (glutathione ,GSH)的调节和再生作用 .方法 :二级SD大鼠 4 0只 ,雌雄各半 ,随机分为 4组 ,为阴性对照组、阳性对照组、低剂量组 (硫辛酸 0 .2g·L-1 ) ,高剂量组(硫辛酸 0 .4g·L-1 ) ,60 Co γ 射线照射剂量为 4Gy .对阳性组、低剂量组、高剂量组动物进行照射后 ,低剂量组和高剂量组动物分别给予硫辛酸油溶液灌胃 (5mL·kg-1 ) .阴性组和阳性组分别给予处理精炼油灌胃作为对照 ;连续灌胃给药 2d后断头处死 ,收集全血静置、析出血清 ,用荧光分光法及分光光度法测定GSH ,MDA和谷胱甘肽过氧化物酶 (glutathioneperoxi dase ,GPx)等指标 .结果 :辐射损伤引起还原型谷胱甘肽含量的降低而氧化型谷胱甘肽 (oxidizedglutathione ,GSSG)的含量增高 (P <0 .0 5 ) ;补充了硫辛酸能增加还原型谷胱甘肽的含量 ,且随着给药剂量的增加而升高 (P <0 .0 5 ) ,GSSG则呈相反趋势 ;硫辛酸能降低辐射损伤造成的GPx的活性的应激性增高 (P <0 .0 5 ) .结论 :硫辛酸对辐射损伤造成的氧化损伤具有保护作用 ,并能调节抗氧化剂GSH的含量 。AIM: To make a model of rats injured by radiation and to find out the effects of lipoic acid on the antioxidation in rats. METHODS: Forty rats were randomly divided into 4 groups: negative control group, positive control group, two trail groups with different dosages of lipoic acid. The dose of lipoic acid was 0.2 g·L -1 and 0.4 g·L -1 in the two trail groups. The rats in the positive control group and two trial groups were radiated by 60 Coγray at the dose of 4 Gy. The two trial groups were given lipoic acid by gavages with different dosages respectively once a day. On the third day after radiation, all the rats were killed and blood was obtained for the analysis of the change of GSH, GSSG, GPX, MDA and protein in blood. RESULTS: After radiation, the content of GSH, GSSG and GPX in positive group was lower than that in the negative group( P <0.05)and the content of GSH and MDA of rats injured by radiation could be elevated by the lipoic acid( P <0.05). CONCLUSION: Lipoic acid can elevate the content of GSH and protect the blood against the damage by radiation.
