摘  要:目的 :了解骨形成蛋白 (Bonemorphogeneticprotein ,BMP)在豚鼠化脓性中耳炎中耳粘膜的表达及定位 ,为研究BMP在慢性中耳炎炎症转归及在鼓室硬化中的作用提供实验依据 .方法 :2 6只豚鼠麻醉后 ,在无菌条件下于右耳经鼓膜注射 1× 1 0 8·L-1 的金葡菌液 1 0 0 μL ,左耳作正常对照 ,制成慢性化脓性中耳炎模型 ,4 5d后用BMP单克隆抗体免疫组化染色对炎症中耳粘膜BMP的表达进行分析 .结果 :中耳粘膜实验组 (n=2 5 )和对照组 (n =2 5 )BMP阳性细胞数分别为 (39.0 4± 5 .34)和 (9.2 4± 1 .83) (t=2 7.1 6 ,P <0 .0 1 ) ,慢性炎症作用下中耳粘膜BMP表达较正常粘膜明显增强 ,阳性细胞主要位于增生小血管周围 .结论 :实验证实BMP在化脓性中耳炎时参与中耳粘膜的炎症反应 。AIM: To explore the expression of Bone Morphogenetic Protein (BMP) in middle ear mucosa of the guinea pig with chronic suppurative otitis media and to supply the experimental data for the role of BMP in the chronic otitis media and tympanosclerosis. METHODS: Twenty six healthy guinea pigs were employed in the experiment. After general anaesthesia, by injecting 1×10 8·L -1 Staphylococcosis aureus into the middle ear cavities of right ears with the left ears serving as controls, the supprative otitis media model was established.Then 45 days later, immunohistochemical ABC method was undertaken to explore the BMP expression in middle ear mucosa of the guinea pigs by using BMP monoclonal antiboby. RESULTS: Number of BMP positive cells in the experiment group ( n =25) and the control group ( n =25) were (39.04±5.34), (9.24±1.83) respectively ( t= 27.16, P <0.01). The presence of inflammation in middle ear induced more BMP expression in the cells than in normal middle ear mucosa cells. The BMP positive cells distributed on the neovasculations.CONCLUSION: BMP possibly plays an important role in the mechanism of otitis media development, and it may play a certain role in the occurrence of tympanosclerosis.
