摘  要:目的 :研究视黄酸调节胸腺细胞分化的作用及其机理。方法 :采用体外胸腺器官培养体系 ,在培养体系中加入视黄酸或受体拮抗剂。在培养的不同时间收集胸腺细胞 ,用荧光标记的抗 CD3、CD4、CD8抗体染色 ,并进行流式细胞仪分析 ,观察胸腺细胞成熟分化的变化。胸腺细胞抽提的 RNA采用实时定量 PCR法检测视黄酸受体 α( RARα) m RNA的表达。结果 :体外胸腺器官培养体系能支持胸腺细胞的发育成熟。 RA在培养的 2 4h促进胸腺细胞向 CD4+分化发育 ,并且该作用可被 RARα特异性拮抗剂 Ro41- 5 2 5 3所拮抗。实时定量 PCR结果显示 ,胸腺细胞表达 RARα m RNA,RA能促进 RARα m RNA的表达 ,并且 RARα m RNA表达水平与 CD4+ CD8+ 胸腺细胞百分数之间存在正相关 ( r=0 .5 93 ,P=0 .0 42 )。结论 :RA对胸腺细胞分化发育具有重要的调节作用 ,RA可能通过影响 RARαmObjective:To study the ability and the mechanisms of RA to modulate thymocyte differentiation.Methods:Thymic organ culture system was used to investigate the role of RA on development of thymocyte. Thymic lobes were cultured in the presence of retinoids and were harvested every day of culture. Thymocyte suspensions were stained with labeled anti-CD3, anti-CD4, anti-CD8 antibody for analysis by flow cytometry. The real time quantitative RT-PCR assay was used to detect the retinoic acid receptor α (RARα) mRNA expression on thymocyte.Results:The thymic organ culture system can support the maturation of thymocyte. Addition of RA to the culture system increased the yield of CD4 + thymocyte, this effect was observed at 24h,but can be antagonisted by Ro41-5253. The expression of RARα mRNA was found in thymocyte. RA can increase the expression of RARα mRNA in thymocyte. A positive correlation was found between the expression of RARα mRNA and the percent of the CD4 +CD8 + cells (r=0.593,P=0.042).Conclusion:RA can regulates thymocyte differentiation, perhaps by affecting the relative expression of RARα mRNA.
