摘 要:以中华绒螯蟹为材料 ,将水体温度从 1 4℃骤然升到 2 8℃ ,然后在升温后 9h内测定中华绒螯蟹肝胰腺中过氧化氢酶 (CAT)、超氧化物歧化酶 (SOD)的活性和丙二醛 (MDA)的含量以及血细胞的吞噬能力 ,比较温度变化对这些指标的影响。结果表明 ,肝胰腺中CAT和SOD的活性在温度升高后马上出现显著降低 ,在 6h后开始趋于稳定 ;MDA的含量在升温后随时间的延长逐步增加 ;血细胞的吞噬力和吞噬指数在温度升高后有所增加并于 3h时达到最高 ,然后逐渐下降到温度升高前的水平。该结果表明 ,温度的骤然升高会影响机体的生理功能和机体的免疫防御能力 ,导致自由基代谢的紊乱 。In order to determine the effect of temperature change on the immune system of the mitten hand crab ( Eriocheir sinensis ), antioxidative and immune parameters were measured within 9 hours of a sudden rise of culture temperature (14℃—28℃). The superoxide dismutase (SOD) in the hepatopancreas was detected by NBT (nitrotetrazolium blue) approach. Compared with the control group, the SOD activity dropped rapidly ( P < 0 05 ) only one hour after the temperature increased. The activity of SOD declined significantly ( P < 0 01 ) and then became stable within 3 hours of the temperature increased. After 6 hours it was still lower than that of the control group. Similar to the SOD, the catalase (CAT) activity also declined suddenly when the temperature increased. The CAT activity declined significantly within 6 hours of the temperature increase then showed stable tendencies, showing no obvious change between 6—9 hours. The concentration of malondiadehyde (MDA) in the hepatopancreas was measured using TBA (2 Thiobarbituric Acid) methods. In contrast to the changes in SOD and CAT , the content of MDA increased after the temperature increase. Within 6 hours of the increase the content of MDA rose significantly ( P < 0 01 ). After 9 hours it continued to rise. The rate and the index of phagocytic both rose, reaching their peak level within 3 hours of the temperature change, before dropping to their original levels. These results indicated that the rapid increment of temperature could result in metabolic maladjustment of free radicals and have profound effects on physiological and immune functions of the body. This could be one of the important ecological factors inducing the outbreak of disease.
