摘 要:目的:研究黄芪总皂甙是否对培养大鼠心肌细胞钙超载产生影响,为研究和开发中药的新成分提供理论依据。方法:异丙肾上腺素干预培养大鼠(1~3d的SD乳大鼠80只)心肌细胞造成心肌细胞钙超载的模型,同时用黄芪总皂甙、维拉帕米和美托洛尔分别作用于细胞72h。分为异丙肾上腺素(isoproterenol,ISO)组,黄芪总皂甙1(astragaosides1,AS1)组,AS2组,ISO+AS1组,ISO+AS2组,ISO+美托洛尔(metoprolol,M)组,ISO+维拉帕米(verapamil,V)组及正常对照组(control,C)组。检测肌细胞内游离钙离子浓度和肌质网的钙负荷,测定心肌细胞内脂质过氧化物丙二醛(maleicdialdehyde,MDA)的含量和超氧化物歧化酶(superoxidedismutase,SOD)的活性。结果:异丙肾上腺素引起心肌细胞游离钙离子浓度和肌质网钙负荷明显增加(分别增加89%和116%)。黄芪总皂甙可浓度依赖性的抑制异丙肾上腺素的作用,从低浓度(10mg/L)到高浓度(100mg/L)。与ISO组比较,ISO+AS1组与ISO+AS2组可使细胞内的钙离子浓度分别降低37%和50%(169.2±24.2),(132.6±29.2),(267.6±82.0)nmol/L,t=2.57,P<0.05;t=3.81,P<0.01,使肌质网钙负荷分别降低23%和35%(P>0.05;t=2.48,P<0.05),但是黄芪总皂甙的这种作用不如维拉帕米,美托洛尔可基本阻断异丙肾上腺素的作用。AIM:To investigate whether the astragalosides can take an effect on calcium o verload in cultured cardiac myocytes of rats, provide the theoretical foundation for the development of the new component of traditional Chinese medicine. METHODS:A model of cardiac myocytes calcium overload was induced by isoproter enol’ s intervention on cultured rats’ (80 SD small rats of 1-3 d)cardiac my ocytes.Cardiomyocytes were exposed to astragalosides,verapamil(V) or metoprolol( M) in the presence of isoproterenol for 72 h respectively,and they were isoprot erenol(ISO) group,astragaosides(AS) group 1,AS group 2,ISO+ AS group 1,ISO+ AS group 2,ISO+ M group,ISO+ V group and control group.Calcium load of free calc ium ions(i) and sarcoplasmic reticulum(SR) in myocyte were detected. Int racellular Maleic Dialdehyde(MDA) and superoxide dismutase(SOD) activity were al so determined. RESULTS:Isoproterenol induced an obvious increase on i and SR calcium load by 89 % and 116 % respectively.Astragalosides had an inhibitory effect to isoproterenol- induced increase oni and SR calcium load with concen tration- dependence, from low concentration(10 mg/L) to high concentration(100 mg/L).Compared with ISO group,ISO+ AS group 1 and ISO+ AS group 2 could lower intracellular calcium concentration by 37% and 50% respectively[(169.2± 24. 2),(132.6± 29.2)vs(267.6± 82.0)nmol/L,t=2.57,P< 0.05; t=3.81,P< 0.01],SR calc ium load decreased by 23% and 35% respectively(P >0.05; t=2.48,P< 0.05). Th e inhibitory effect of astragalosides was less than that of verapamil.Metoprolol almost inhibited completely isoproterenol- induced increase on i and S R calcium load. In addition,ISO increased intracellular MDA content(t=2.57,P < 0 .05),astragalosides had an decreasing tendency to isoproterenol- induced increa se on MDA, and had an increasing tendency in SOD activity. CONCLUSION:Astragalosides can inhibit calcium overload of cardiac myocytes in duced by isoproterenol, one of the mechanisms is to reduce SR calcium load and p revent lipid peroxidation.
