摘 要:目的探讨低频低强度超声对超广谱β-内酰胺酶(ESBLs)大肠杆菌外膜通透性的影响,以及其联合抗生素对产ESBLs大肠杆菌的协同增效杀菌作用。方法抗生素选用庆大霉素和妥布霉素,实验分为对照组、药物组、超声组、联合组(即超声联合药物)。实验前后对菌液进行细菌计数、电镜观察、碘化丙啶(PI)荧光值检测。结果 (1)细菌生存率随超声辐照时间延长出现显著下降。(2)透射电镜显示低频低强度超声能改变细菌外膜的通透性。(3)辐照后,细菌死亡率与外膜通透性率呈正相关。结论低频低强度超声联合抗生素对ESBLs大肠杆菌的协同增效杀菌作用显著,且能改变其通透性。Objective To study the effect of low-frequency and low-intensity ultrasound on permeability of the outer membrane and the synergistic effect of ultrasonic irradiation combined with antibiotics against the vitality of Extended-spectrum Beta-lactamases(ESBLs)producing Escherichia coli.Methods The standard suspension were randomly divided into 4groups:the control group,the drug group,the ultrasonic irradiation group and the combination of drug and ultrasonic irradiation group.Gentamicin and tobramycin were the therapeutic drug.The vitality and the permeability of the bacteria of each groups were measured after the experiment processing.Results The vitality of the suspension experienced a significant reducing after experimental treatment.The fluorescence intensity of Propidium Iodide(PI)and the Transmission Electron Microscopy(TEM)showed the permeability of the bacteria was enhanced after ultrasonic irradiation.Conclusions A certain dose of ultrasound may change the permeability of ESBLs-producing Escherichia coli and it’s synergistic effect of ultrasonic irradiation with antibiotics against ESBLs-producing Escherichia coli is also conspicuous.
