摘  要:目的鉴定自鸵鸟粪便内分离到的隐孢子虫虫种,了解其生物学特性。方法显微镜观察卵囊形态结构,基于18SrRNA基因位点进行PCR扩增。建立试验动物感染模型,通过临床症状、排卵囊规律和组织学等方面进行观察研究。结果卵囊大小为(7.41±0.12)μm×(5.70±0.07)μm,卵囊指数为1.30,经序列分析鉴定该分离株为鼠隐孢子虫(Cryptosporidium muris)。Balb/c鼠和蒙古沙鼠接种1.0×106个鸵鸟源C.muris卵囊后,潜隐期分别为9d和13d,显露期分别为37d和72d,排卵囊高峰期分别在接种后17~31d和接种后31~64d;罗曼雏鸡不感染鸵鸟源C.muris。感染C.muris的试验动物均未表现出胃肠道紊乱的临床症状。组织学和电镜扫描观察发现大量虫体寄生于胃腺粘膜内,并可见其各发育阶段。结论分离鉴定鸵鸟源C.muris,成功建立Balb/c鼠和蒙古沙鼠感染模型,为隐孢子虫的分类地位及人兽共患性研究提供了参考。We identified Cryptosporidiumspecies in ostriches,and studied the biological characterization of the Cryptosporidiumisolate.The morphology of oocysts were observed by microscope,and examined by PCR amplification of 18 srRNA gene.Experimental infections were established,clinlcal symptoms,pattern of shedding oocysts and histological examination were observed.Results showed that the oocyst measured 7.41±0.12μm × 5.70±0.07μm,with a length/width ratio of1.30,and the isolate was conformed as Cryptosporidium murisby DNA sequence analysis.The 1×106oocysts of C.muris were inoculated to BABL/c mice and Mongolian gerbil,with the prepatent period of 9days(d)and 13 d,the patent period of 37 d and 72 d,the peak of parasitization around 17-31 days post-infection and 31-64 days post-infection,respectively;chickens were not infected with C.muris derived from ostrich.The experimental animals of infected with C.muris did not showed clinical symptoms of gastrointestinal disturbance.Histological and electron microscopy examination found that a large number of parasites were located in the gastric mucosa and the most endogenous stages were observed.In this study,C.muris were identified in ostriches,and experimental model have been established in BABL/c mice and Mongolian gerbil,providing useful reference for taxonomy and zoonotic of Cryptosporidium.
