RP-HPLC同时测定心安宁片中葛根素和2,3,5,4'-四羟基二苯乙烯-2-O-β-D-葡萄糖苷的含量

摘　　要：目的:建立心安宁片中葛根素和2,3,5,4’-四羟基二苯乙烯-2-O-β-D-葡萄糖苷含量的测定方法。方法:采用反相高效液相色谱法,以Nova-Pak C18(5μm,150 mm×4.6 mm)柱为分析柱,乙腈-水(15:85)为流动相,流速:0.8 mL·min-1,检测波长:320 nm,柱温:40℃,以外标法检测。结果:葛根素在29.76～297.60μg·mL-1浓度范围内线性关系良好(r=0.9999),平均回收率为101.2%,RSD为1.6%(n=6);2,3,5,4’-四羟基二苯乙烯-2-O-β-D-葡萄糖苷在1.824～18.24μg·mL-1浓度范围内线性关系良好(r=0.9992),平均回收率为99.32%,RSD为2.2%(n=6)。结论:该法快速、简便,分离度好,可同时测定心安宁片中葛根素和2,3,5,4’-四羟基二苯乙烯-2-O-β-D-葡萄糖苷的含量。Objective:To establish a method for the determination of puerarin and 2,3,5,4' - tetrahydroxy - stibe-ne -2 - O -β- D - glycoside in Xin' anning tablets. Methods: RP - HPLC was conducted on a Nova - Pak C18 column (5μm ,150 mm × 4. 6 mm) with acetonitrile - water (15: 85 ) , flow rate 0. 8 mL· min-1, detecting at 320 nm and column temperature at 40 ℃. The external standard method was adopted for the assay. Results: The linear was 29. 76 -297. 60 μg· mL-1 (r =0. 9999) ,the average recovery was 101. 2% ,RSD = 1. 6% (n =6)for puerarin and the linear was 1. 824 - 18. 24μg· mL-1 (r =0. 9992) ,the average recovery was 99. 32% ,RSD =2. 2% (n = 6 ) for 2,3,5,4' - tetrahydroxy - stibene -2-O-β-D- glycoside. Conclusions: The method is simple, sensitive, accurate and suitable for the determination of puerarin and 2,3,5,4' - tetrahydroxy - stibene -2-O-β-D-glycoside in Xin' anning tablets.
