摘  要:目的:研究毛翠雀提取物对博莱霉素(BLM)所致实验性大鼠肺纤维化的干预作用,并初步探讨其作用机制。方法:SD大鼠60只,随机分为6组:正常对照组、模型组、醋酸泼尼松(3.5 mg/kg)阳性对照组及毛翠雀水提物高(4 g/kg)、中(2 g/kg)、低(1 g/kg)剂量组。除正常对照组外,其余各组大鼠采用气管内一次性注射盐酸博莱霉素复制肺纤维化模型,正常对照组滴注等体积生理盐水。造模后第2天开始灌胃给药,1次/d,连续14 d。正常对照组和模型组给予等体积生理盐水。给药14 d后处死大鼠,分别检测大鼠肺系数及血清羟脯氨酸(HYP)、总抗氧化能力(T-AOC)、还原性谷胱甘肽(GSH)、丙二醛(MDA)、超氧化物歧化酶(SOD)、肿瘤坏死因子(TNF-α)、生长转化因子(TGF-β_1)、白介素1β(IL-1β)、白介素4(IL-4)、白介素10(IL-10)等指标。同时取固定部位肺组织分别行HE、Masson染色,进行组织病理学观察。结果:与模型组比较,毛翠雀提取物能显著降低肺纤维化大鼠的肺系数及血清HYP、MDA、IL-1β、TNF-α水平,升高血清GSH、T-AOC、SOD、IL-4、IL-10、TGF-β_1水平(P<0.05或P<0.01),明显改善肺泡炎症和肺纤维化程度。结论:毛翠雀对博莱霉素所致实验性肺纤维化大鼠有一定的保护作用,其机制可能与抗氧化损伤和抑制炎症细胞的浸润活化有关。Objective:To observe the influence of extracts of Delphinium trichophorum on bleomycin(BLM)-induced pulmonary fibrosis in rats.Methods:60 SD rats were randomly divided into six groups,normal group,model group,prednisone acetate group(3.5 mg/kg)and Delphinium trichophorum high(4 g/kg)-,medium(2 g/kg)-,low(1 g/kg)-dose group.In addition to the normal group,the rats in other groups were treated with intravenous injection of bleomycin hydrochloride to replicate the pulmonary fibrosis model,and the normal group was injected with saline.The medicine was given on the second day after the molding,1 time/d,and continuous for 14 d.Normal group and model group were given equal volume physiological saline.After 14 d,rats were killed.The levels of rat lung coefficient,HYP,T-AOC,GSH,MDA,SOD,TNF-α,TGF-β1,IL-1β,IL-4 and IL-10 in lung tissues were measured and analyzed,then the lung tissue of fixed parts were obtained by HE and Masson staining for histopathological examination.Results:Compared with the model group,the levels of HYP,MDA,IL-1βand TNF-αdecreased,and the expressions of GSH,T-AOC,SOD,IL-4,IL-10 and TGF-β1 in lung tissues were dramatically increased(P<0.05 or P<0.01)after Delphinium trichophorum extracts treatment.Pulmonary alveolar inflammation and pulmonary fibrosis were significantly improved.Conclusion:Delphinium trichophorum has a certain protective effect on the experimental pulmonary fibrosis rats which induced by the bleomycin.The mechanism may be related to the antioxidation injury and the infiltration activation of inflammatory cells.
