脂质体包裹人血红蛋白微囊的制备及其对大鼠血液的影响

摘　　要：目的 :探讨用人血红细胞经低渗破膜释放出血红蛋白 (Hb) ,将Hb经过离心、超滤并制成血液代用品的新途径。方法 :采用脂质体包裹Hb的方法 ,并将制备脂质体包裹血红蛋白 (LEH)通过大鼠股静脉换血试验输入体内 ,异侧股动脉放等体积血 ,检测受试大鼠血液中的血红蛋白、红细胞、白细胞及血小板。结果 :制得Hb平均浓度为141g/L(5批 )、纯度为97.7 %稳定的LEH ,LEH包裹率为27.7% ,冰冻保存泄漏率<10 % ,LEH的直径为(237.5±37.3)nm。结论 :LEH是具有生物学活性的新型载氧制剂。Objective:To study a new way to produce aritificial red blood ce lls with human hemoglobin by centrifugation and ultrafilter.Methods:The Hb was centrifuged,ultra_filtered and encapsulated with lipoˉsome.Results:Average concentration of acquired Hb was141g/L(5lots),and its purity was97.7%with stable liposome.Packing rate of LEH was27.7%.The diameter of LEH was(237.5±37.3)nm,and the frozen storage leakiness was less than10%.LEH was transfused into rat through thigh vein,and the same volume of blood was released from the other thigh artery.The Hb,RBC,WBC,PLT of rat were detected.Conclusion:LEH had bioˉlogical activity,and was a new lower immunogenic product for carrying oxygen.
