新的荚膜染色方法

摘　　要：将感染Ⅲ型肺炎球菌的小白鼠,取心、肺、肝等内脏组织的血液,推成厚薄适中的血膜,自然干燥。甲醇固定,吹干。革兰氏第一液染色。20%Cuso_4水溶液冲去染液。革兰氏第二液作用。结果:荚膜染成淡兰色,荚膜周围围一个内外边沿整齐的褐色堤状浓染区。菌体呈深兰色,红血球呈淡黄色。背景呈均匀一致且很鲜艳的天兰色。荚膜对比非常清晰。The white rate was infected by pneunococcusⅢ.It is essential to spread a appropriate film of the specimen on to a glass slide with the mouse blood from heart,lung or liver, then drying naturally,fixed with methanol,again and then air dry.Thefirst stainng is Gram's Ⅰ dye liguid which is added to specimen,then the dye liguid is washed off with 20% Cuso_4,then adding Gram's Ⅱ.As a resulf of staining,the capsules became light blue.A broun sediment is formed round capsules.The bodies turned dark blue.The red blood cell is light yellow,The back ground is well-distributed sky-blue.The cupsules are very clear.
