珍珠梅活性成分的提取分离及体内抑瘤作用的研究

摘　　要：目的 :对珍珠梅进行化学成分和体内抑瘤作用的研究。方法 :用硅胶柱色谱法对珍珠梅乙酸乙酯提取物进行分离纯化 ,通过理化常数和光谱分析鉴定化合物结构 ;并按照标准方法给 36只小鼠接种S180 肉瘤后 ,灌胃给予珍珠梅乙酸乙酯提取物 10d ,观察瘤重及体重 ,检测NK细胞活性及血清肿瘤坏死因子 (TNF α)、白介素 2 (IL 2 )的含量。结果 :从珍珠梅乙酸乙酯提取物中分离 3种化合物 ,分别鉴定为 :5 ,2′ ,4′ 三羟基 6 ,7,5′ 三甲氧基黄酮 (Ⅰ ) ,丁二酸 (Ⅱ ) ,对羟基苯甲酸 (Ⅲ ) ;实验组与对照组瘤重有显著性差异 (P <0 0 5 ) ,实验组与对照组体重变化无显著性差异 (P >0 0 5 ) ,实验组脾细胞NK细胞活性及血清TNF α、IL 2的含量明显高于对照组 (P <0 0 5或P <0 0 1)。结论 :化合物Ⅰ、Ⅱ为首次从珍珠梅中获得。珍珠梅乙酸乙酯提取物可以改善S180 荷瘤小鼠免疫功能 ,增强机体抗肿瘤能力。Objective: To study the constituents from Sorbaria sorbifolia and their antitumor activities. Methods: The constituents were isolated with silia gel column chromatography and identified by physicochemical properties and spectroscopic analysis. Thirty six mice were inoculated with sarcocarcinoma 180 according to the standard method, Sorbaria sorbifolia ethyl acetate etract was administrated for 10 days, tumor and body weight, the activities of NK cell and the contents of TNF-α, IL-2 in serum were detected. Results: Three compounds were isolated from the ethyl acetate extract and identified as 5,2′,4′-trihydroxy-6,7,5′-trimethoxyflavone(Ⅰ),succinic acid(Ⅱ) and p-hydroxybenzoic acid(Ⅲ). The tumor of the test groups were lighter than that of control group(P<0.05). There was no remarkable difference between the control and test groups in body weight (P>0.05). The activitiy of NK cell and the contents of TNF-α,IL-2 in serum were higher than that of control group (P<0.05). Conclusion: Compounds Ⅰ and Ⅱ were isolated from Sorbaria sorbifolia ethyl acetate extract for the first time. The ethyl acetate extract could improve immune function and strengthen antitumor effect in sarcocarcinoma 180.
