摘 要:目的 研究大鼠小肠 (十二指肠、空肠、回肠 )和大肠 (乙状结肠 )上皮内淋巴细胞 (IEL)、固有层内淋巴细胞 (LPL)分布特点。方法 通过免疫组化双重染色 ,标记肠绒毛基底膜 ,区分出上皮和固有层 ,然后分别对IEL和LPL计数 ,并进行统计学分析。结果 大鼠肠道各部分 (十二指肠、空肠、回肠、乙状结肠 )IEL数量依次递减 ;与此相反 ,LPL数量依次递增。而且它们的T细胞亚群 (CD3、CD4、CD8、TCRαβ、TCRγδ)数量也存在显著性差异 (P <0 .0 5 )。结论 通过双重染色标记出基底膜来计数IEL、LPL的方法非常精确 ,肠道各段的IEL。Objective To study the distributed features of intraepithelial lymphocyte(IEL) and lamina propria lymphocyte(LPL) of small intestine(duodenum,jejunum,ileum) and large intestine(sigmoid colon) in rat.Methods Basilar membrane of bowel villi was marked by immunohistochemistry double staining,differentiate epithelia and lamina propria,and the amount of IEL and LPL were counted.Results The amount of IEL in various parts(duodenum,jejunum,ileum and sigmoid colon) of rat intestinal tract decreased in turn.On the contrary,the amount of LPL increased in turn.The difference among T cell subsets were obvious(All stated differences achieved statistical significance at P ≤0.05).Conclusion The method of double staining to account the number of IEL and LPL is very accurate,and IEL and LPL in different part of the intestine tract may play different biological roles in immune response of mucosa.
