摘 要:观察CHO C2 8细胞在大生产过程中 ,生长状态曲线 ,为细胞培养过程中换液及培养条件提供依据。人为控制培养液中血清含量、加液量、pH值等条件。从生产种子 2 2代左右开始传代 ,经 5~ 6 d形成致密细胞单层 ,一般维持换液次数为 30次左右。如果生产种子代次高 ,其维持换液次数也相应减少 ,根据不同换液次数细胞增殖数量不同 ,总结出重组CHO C2 8细胞生长动态曲线。并根据换收液的HBsAg滴度 ,绘出换液次数和HB sAg相关曲线。结论 :7~ 2 3次换液期间 (缓慢生长期 )HBsAg收获最高。To observe growth curve of CHO-C 28 cell in large-scale production, and to provide essential condition for replacing medium and culture conditions in cell culture. When cells have formed compact layer in 15L roller bottle, change culture medium every two days, and control serum content, medium volume, pH. The twenty-second generation cell was passaged and form compact layer after five to six days. The times of changing media may keep about thirty times. If seeds are older, the times of changing media will be correspondingly reduced. According to cell amounts after changing medium, draw growth dynamic curve of recombinant CHO-C 28 cell. According to titer of HBsAg, draw the curve of the times of changing media and HBsAg. In the seven to twenty-three times change medium period (slow growth period), the harvest of HBsAg reached the larger quantity.
