四逆散及其各组成中药对实验性肝损伤的影响  被引量:13

摘  要:目的 :对四逆散进行初步拆方研究 ,探讨组成四逆散的各单味中药在全方中所起的作用。方法 :运用CCl4体外致肝细胞损伤及 2 ,4 ,6 三硝基氯苯 (PCl)致迟发型变态反应 (PCl DTH)诱导肝损伤模型 ,ConA致淋转试验 ,LPS诱导巨噬细胞释放NO试验及活化脾细胞释放IV胶原酶能力试验 ,考察四逆散全方及各单味中药对肝细胞及免疫细胞的影响。结果 :四逆散复方作用明显优于各单味中药 ;而各单味药分别作用于肝损伤过程的不同环节 ,柴胡主要通过保护肝细胞膜及促进肝保护因子NO的产生而影响肝损伤过程的主要环节 ,白芍则主要抑制免疫细胞的活化及过度活化的免疫细胞的功能而发挥免疫调节作用 ,甘草具有保肝、促进NO产生及免疫调节作用。结论 :四逆散全方作用优于各单味中药 ,系各单味中药对疾病过程中不同靶点综合效应的结果。AIM: To examine the function of the four constituent medicines of Si Ni San(Composition of Radix Bupleuri,Radix Paeoniae alba, Fructus Aurantii immaturus and Radix Glycyrrhizae) and each single drug on alleviating experimental liver injury METHOD: Hepatocyte damage induced by CCl 4 and PCl DTH,Con A induce lymphocyte proliferation,LPS induced NO production in macrophage and gelatin zymography assay were used RESULT: The results show that the activities of Si Ni San were stronger than those of each constituent Among the four constituent medicines,Radix Bupleuri mainly affected the main process of liver injury through protecting hepatocyte membrane and enhancing NO,a protective factor,production in macrophages;Radix Paeoniae alba mainly affected immune cells through inhibiting the activation,migration and infiltration of lymphocytes,and inducing the dysfunction of liver infiltrating cells; Radix Glycyrrhizae had both hepatoprotective and immunomodulatory activities contributing to SNS alleviating liver injury CONCLUSION: Each constituent of Si Ni San affects the different target of liver injury and their comprehensive effect resulted in the stronger effect of the prescription
