妊高征胎盘滋养细胞中IGF-ⅡmRNA的表达及意义

摘　　要：目的　检测滋养细胞中胰岛素样生长因子ⅡmRNA(IGF -ⅡmRNA)表达水平 ,评估IGF -Ⅱ在妊娠高血压综合征 (妊高征 )中的作用。方法　用逆转录 -聚合酶链反应 (RT -PCR)检测正常妊娠滋养细胞 15例、妊高征滋养细胞 15例中IGF -ⅡmRNA表达水平 ,并通过紫外凝胶图像分析进行定量分析比较。结果　与正常滋养细胞比较 ,妊高征滋养细胞IGF -ⅡmRNA的表达强度显著降低 (t =2 .38,P <0 .0 5 )。结论　滋养细胞表达IGF -Ⅱ的多少 ,与妊高征的发生有关。Objective: Quantitatively determining insulin-like growth factor-Ⅱ mRNA (IGF-Ⅱ mRNA) in trophoblasts by reverse transcription-polymerase chain reaction(RT-PCR), and to evaluate the role of IGF-Ⅱ in pregnancy induced hypertension syndrome(PIH). Methods: Control group was consisted of fifteen normal pregnancies. Study group was consisted of fifteen women with PIH. IGF-Ⅱ mRNA expression in trophoblasts was detected by RT-PCR. The mean density was determined by image analysis technique. Results: The expression level of IGF-Ⅱ mRNA of PIH group was significantly lower than that of control group. Conclusion: These results suggested that IGF-Ⅱ mRNA in human placenta was principally expressed in trophoblast. IGF-Ⅱ has the improtant role for PIH forming.
