泛影葡胺对糖尿病肾病大鼠心肌细胞内皮素受体A表达的影响

摘　　要：目的 :观察造影剂泛影葡胺对糖尿病肾病大鼠心肌细胞内皮素受体A (endothelinreceptorA ,ETAR)表达的影响。方法 :应用链脲佐菌素 (STZ)腹腔注射建立SD大鼠糖尿病肾病模型。设对照组、模型组、泛影葡胺组 ,BQ1 2 3 +泛影葡胺组 ,利用RT PCR和westernblot方法检测心肌组织ETAR表达的变化。结果 :与对照组比较 ,各组糖尿病大鼠心肌ETAR表达上调 ,CM组上调尤为明显(P <0 .0 5) ,注射BQ1 2 3后 ,可见其表达下调。结论 :有糖尿病肾病基础病史时 ,泛影葡胺可引起心肌组织ETAR表达变化 ,从而影响心脏功能 ;预防性使用选择性ETAR拮抗剂BQ1 2 3 ,可下调ETAR蛋白表达 。AIM: To investigate the effects of diatrizoate on the expres si on of endothelin receptor A (ETAR) in cardiac myocyte of diabetic nephropathy SD rats. METHEDS: 32 male SD rats (220 -280 g) were randomly divid ed into four groups: 8 normal control rats (Con group) with sham surgeon operati on; the other 24 were diabetic rats which were induced by intraperitoneal strept ozotocin ( 35 mg·kg -1 body weight) at one dose after they were unineph rectomized for 2 weeks. At the end of the eighth week, the diabetic rats were di vided into 3 groups at random: (DM group); diabetic rats plus diatrizoate inject ion ( 10 ml·kg -1 ) through tail vein of rats (CM group); BQ123 treated group (BQ group) which were injected the BQ123 ( 1 mg·kg -1 ) before bei ng injected the diatrizoate, while the Con group were only given the sterile wat er. ETAR was examined by reverse transcription polymerase chain reaction (RT-PC R) and western blot. RESULTS: Both RT-PCR and Western blot analysis indicated that ETAR had been upregulated in CM, DM and BQ groups compa red with that in Con group (P< 0.05 ); especially in CM group . BQ123 could downregulate the expression of ETAR gene induced by diatrizoate. CONCLUSION: Increasing ETAR may affect cardiac function induced by radiocontrast media diatrizoate under diabetic nephropathy conditions. Howeve r, BQ123 can downregulate the expression of ETAR gene induced by radiocontrast m edia diatrizoate and improve cardiac function.
