摘  要:目的:探讨糖尿病时肝组织胰岛素样生长因子-1(IGF-1)基因表达的异常及其与糖尿病外周神经病变的关系。方法:分离解剖出右侧坐骨神经,测定诱发电位出现的波幅(amplitudeofevokedpotentials,AEP)、感觉及运动神经传导速度(sensory/motornerveconductionvelocity,SNCV/MNCV),用四氧嘧啶诱发糖尿病SD大鼠模型。48只糖尿病大鼠按经(胰岛素)控制后血糖水平分成3组:ID-1,2,3组,16只正常大鼠用作对照组。反转录一多聚酶链扩增反应半定量分析坐骨神经IGF-1mRNA含量;酶联免疫吸附法分析组织IGF-1肽含量;诱发肌电图测定坐骨神经电生理指标;观察坐骨神经形态学改变。结果:病程早期(2周,2个月),正常对照组、ID-2组大鼠肝组织IGF-1mRNA含量(1.15±0.090,0.79±0.048,P<0.001;1.17±0.069,0.53±0.023,P<0.001)、肽含量均下降犤(196.66±14.9),(128.2±11.25)μg/g,P<0.001;(196.66±14.9),(74.43±5.33)μg/g,P<0.001犦出现在相应糖尿病组大鼠坐骨神经电生理指标异常之前,程度均随糖尿病控制状态而异,且随病程进一步逐渐下降犤IGF-1mRNA(0.71±0.024)~(0.47±0.021);IGF-1肽(114.35±8.09)~(64.58±3.89)μg/g,P<0.001犦。血清IGF-1呈一致性下降(r=0.99,P<0.001)。其变化与坐骨神经功能改变(感觉神经:r=0.54,P<0.05,AIM:To explore the relation between insulin like growth factor 1 (IGF 1) gene expression abnormality of liver tissue and pathologic changes of peripheral nerves in diabetes. METHODS:Diabetes was induced in Sprague Dawley(SD) rats by alloxan. Diabetic rats were then subdivided into three groups of 16 rats according to glucose level after control by insulin:insulin controlled diabetes 1,2,3 group.Sisteen normal rats were used as the normal control group.Their sciatic nerves were isolated.The latency,amplitude of evoked potentials(AEP) and the sensory/motor nerve conduction velocity(SNCY/MNCV) were measured with Viking IV evoked electromyography.One gram of liver tissue was removed,the total RNA was extracted The IGF 1 mRNA was determined semi quantitatively by using the reverse transcription polymerase chain reaction(RT PCR),the content of IGF 1 peptide inats according to glucose level after control by insulin: insulin controlled diabetes 1,2,3 group.Sisteen normal rats were used as the normal control group.Their sciatic nerves were isolated.The latency,amplitude of evoked potentials(AEP) and the sensory/motor nerve conduction velocity(SNCY/MNCV) were measured with Viking IV evoked electromyography.One gram of liver tissue was removed.The total RNA was extracted The IGF 1 mRNA was determined semi quantitatively by using the reverse transcription polymerase chain reaction(RT PCR).The content of IGF 1 peptide in liver tissue and that in the serum were determined by enzyme linked immunosorbent assay(ELISA).The morphology of sciatic nerves was observed. RESULTS:In the 2nd week of disease course IGF 1 mRNA content in liver tissue was (1.15± 0.090) in the control group and(0.79± 0.048) in the ID 2 group(P< 0.001);in the 2nd month of disease course,IGF 1 mRNA content was(1.17± 0.069) in the control group and(0.53± 0.023) in the ID 2 group(P< 0.001).In the 2nd week of disease course the IGF 1 peptide content was(196.66± 14.9) μ g/g in the control group and(128.2± 11.25) μ g
