几种酶在大鼠内吞体膜上的细胞化学定位

摘  要:目的研究多种酶在不同器官内吞体膜上的活性。方法用细胞化学方法,以Ce3+或Pb2+作捕捉剂,分别检测碳酸酐酶,Mg2+-ATP酶,Na+-K+-ATP酶,5’-核苷酸酶,NADPH oxidase在大鼠肾近曲小管上皮细胞、肝细胞以及人体嗜中性白细胞内吞体膜上的酶活性。结果上述碳酸酐醇,Mg2+-ATP酶,Na+-K+-ATP酶,5’-核苷酸酶及NADPH oxidase分别在大鼠肾脏近曲小管,肝脏及人体白细胞的内吞体膜上都有酶活性存在。结论 内吞体功能复杂,它可能与细胞的pH值,水盐代谢,能量、蛋白质的代谢以及活性氧的产生等都有一定的关系。Objective To investigate cytochemical localization of enzymes on the membrane of endosome in different organs. Methods To use Ce3+ or Pb2+ as capture agents to detect carbonic anhydrase, Na+ - K+ - ATPase, Mg2+ -ATPase, 5'-nucleotidase and NADPH oxidase activities on endosome membrane in epithelial cells of renal proximal tubules,hepatic cell of rat and human leukocytes, with cytochemical method. Results Carbonic anhydrase,Na+ -K+ -ATPase ,Mg2+-ATPase,5'-nucleotidase and NADPH oxidase could manifest enzyme activity on endosome membrane in the renal proximal tubules, hepatic cell of rat and human leukocytes. Conclusion Endosome has complicated function and probably relates with cellular pH,the metabolism of water and salt, the metabolism of energy , protein and the production of active oxygen.
