猪胚胎细胞核移植重组胚发育能力的研究

摘  要:选用香猪 2 ~ 16 细胞期卵裂球作核供体 ,上海白猪卵母细胞作核受体。通过显微操作和电融合技术构成重组胚。体外培养时 ,以融合前 1h激活、融合前 2h激活及融合前不激活的卵母细胞作核受体的重组胚 ,发育率分别为 6 5 .8% ,5 3.1% ,33.3%。以体内、体外成熟卵母细胞作核受体胞质时 ,其重组胚的发育能力无显著变化 (P >0 .0 5 )。作为核供体的胚胎对于重组胚的发育能力有显著影响 ,母体合子转变以后的胚胎作为核供体时 ,重组胚的发育能力下降 ;分别以不同细胞期胚胎 (2 ,4 ,8 和 16 细胞 )的卵裂球作为核供体的NT胚发育率有显著差异 (P <0 .0 5 ) ,以 2 ~ 4 细胞胚胎卵裂球作核供体最合适。cell embryos blastomere of Xiang pig were used as nuclear donor and Shanghai White pig used as nuclear recipient. Reconstructive embryos were produced with electrofused method by micromanuplantation. The developmental rate of nuclear transfer embryos(NTE) was 65.8%, 53.1% and 33.3% respectively, when the recipient oocytes were activated two hours, one hour prior to fusion and nonactivated. The developmental rate of NET was 58.8% and 64.0% (P>0.05)respectively, showed no significant difference when the recipient oocytes matured in vivo and in vitro. The developmental rate of NET(>4-cell) was 51.0%,48.3%,38.9% and 21.6% respectively,when 2-,4-,8-,and 16-cell embryos were used as donor embryos. Therefore,2-~16-cell embryos blastomere were more suitable for nuclear donor.
