摘 要:目的 探讨硫酸锌盐对体外培养的人视网膜色素上皮(retinal pigment epithelium,RPE)细胞增生及Caspase-3表达的影响。方法 采用MTT法检测不同浓度硫酸锌盐对RPE细胞生长的影响,用TUNEL技术检测RPE细胞凋亡,免疫细胞化学染色观察Caspase-3表达。结果 锌盐浓度高于0.001μmol/L就可抑制RPE细胞的增生;当锌盐浓度高于10μmol/L后对RPE细胞的增生抑制率与药物剂量成正比。不同浓度锌盐均可诱导RPE细胞表达Caspase-3增加,且当锌盐浓度高于0.01μmol/L就可诱导RPE细胞凋亡。结论 锌盐可诱导RPE细胞表达Caspase-3增加,从而可引发RPE细胞的凋亡。因此对于不缺乏锌的年龄相关性黄斑变性(age-related macular degen-eration,ARMD)患者锌剂治疗未必有益。Objective To clarify the effects of zinc on the proliferation and the expression of caspase-3 in cultured human retinal pigment epithelial (RPE) cells. Methods The effects of zinc on the proliferation of RPE were measured by MTT method. TUNEL method was used to detect the apoptosis of RPE cells. The expression of caspase-3 was delected by immunohistochemistry. Results A concentration of zinc over 0. 001μmol/L could inhibit the proliferation of RPE. The relationship between concentration of zinc more than 10μmol/L and growth prohibition rate of RPE cells was dose-dependent. All concentrations of zinc including 0. 001μmol/L enhanced the expression of caspase-3 of RPE. The concentrations of zinc more than 0. 01μmol/L could induce apoptosis of RPE. Conclusion Zinc could induce apoptosis and enhance the expression of caspase-3 of RPE cells. Caution should be taken when using zinc supplements for the treatment of ARMD patients without deficiency of zinc.
