摘 要:目的通过借助经典光毒剂8-甲氧补骨酯素(8-MOP)研究UVA、UVB、UVA+UVB3种不同光源对光毒性斑贴试验结果的影响。方法豚鼠138只,健康受试者22名。光源为PhilipsUVA,PhilipsUVB,SUV1000日光模拟器。试验物为2%8-MOP。①豚鼠对8-MOP+UVA(PUVA)和8-MOP+UVB(PUVB)的反应。②豚鼠对8-MOP+UVA(PUVA)和8-MOP+UVA+UVB(PUVA+UVB)的反应。③人体对8-MOP+UVA(PUVA)和8-MOP+UVA+UVB(PUVA+UVB)的反应。结果PUVA组的反应率和单纯UVA照射组有显著差异(P<0.01),PUVB组的反应率和单纯UVB照射组的差异无显著性(P>0.05)。PUVA+UVB组试验区域3d皮肤反应评分变化趋势的斜率大于PUVA组。人体试验结果同动物试验结果相一致。结论在以8-MOP为受试物的光斑贴试验中,UVB检测效果较差,而UVA和PUVA+UVB均能检测出8-MOP的光毒性,但UVA+UVB要优于UVA。因而UVA+UVB是一种理想的光斑贴试验光源。Objective To compare the results of8-methoxypsoralen(8-MOP)to skin reaction by photo-patch test with ultraviolet-A(UVA),ultraviolet-B(UVB)and UVA+UVB.Methods A total of138healthy guinea pigs and22healthy volunteers were included;the light source were Philips UVA,Philips UVB,SUV1000sunlight simulator.Concen-tration of8-MOP was2%.All the subjects were divided into3groups:①Guinea pigs reaction to8-MOP+UVA(PUVA)and8-MOP+UVB(PUVB);②Guinea pigs reaction to8-MOP+UVA(PUVA)and8-MOP+UVA+UVB(PUVA+UVB);③Volunteers reaction to8-MOP+UVA(PUVA)and8-MOP+UVA+UVB(PUVA+UVB).Results The reaction rate between group PUVA and UVA was variant significantly(P<0.01),while there was no significant difference between PUVB group and UVB group(P>0.05).In PUVA+UVB testing area in guinea pigs the3-day skin reaction grade was great than that of PUVA testing area,the results in volunteers were basically the same as that of the guinea pigs.Conclusion In8-MOP photo-patch test,UVB could not detect the phototoxicity of8-MOP,while UVA and PUVA+UVA could both detect the phototoxicity of8-MOP,and UVA+UVB is superior to UVA.UVA+UVB sould be an ideal a choice for photo-patch test.
