摘  要:目的 研究白介素 1β(interleukin 1,IL 1β)对正常皮肤、增生性瘢痕和瘢痕疙瘩中的成纤维细胞的表皮生长因子 (EGF)、表皮生长因子受体 (EGFR)、转移生长因子 βR1(TGFβR1)的作用。方法 将体外培养 5代之内的 6例瘢痕疙瘩、6例增生性瘢痕及 6例正常皮肤的成纤维细胞分别置于含IL 1β( 2 0 0U/ml)的 10 %胎牛血清DMEM培养液中 ,孵育 3d ,免疫细胞化学染色显示EGF、EGFR、TGFβR1。结果 正常皮肤、增生性瘢痕、瘢痕疙瘩各组中成纤维细胞的EGF相对含量分别为 162 .0 0±2 0 .79;15 1.0 0± 2 6.45 ;147.5 0± 2 3 .61。EGFR含量分别为 148.3 4± 13 .72 ;13 6.67± 17.5 2 ;13 1.67±11.71。TGFβRI 含量 113 .5 2± 17.62 ;10 6.71± 2 1.43 ;10 7.5 4± 2 3 .5 2。三者成纤维细胞EGF、EGFR和TGFβRI 含量差异无显著性 (P >0 .0 5 )。加入IL 1β后 ,成纤维细胞EGF、EGFR和TGFβRI 相对含量 :正常皮肤为 2 2 4.0 0± 3 1.5 9、178.67± 2 7.86和 80 .3 4± 11.75 ;增生性瘢痕为 12 8.75± 18.3 1、10 5 .82± 2 1.61和 10 9.83± 2 5 .79;瘢痕疙瘩 113 .0 1± 2 4.71、93 .3 4± 3 0 .71和 118.75± 19.2 7。正常皮肤成纤维细胞EGF和EGFR相对含量明显高于增生性瘢痕和瘢痕疙瘩 (P <0 .0 5 ) 。Objective To investigate the effect of interleukin-β1(IL-1β) on epidermal growth factor (EGF), epidermal growth factor receptor (EGFR) and transforming growth factorβR 1 (TGFβR 1) in fibroblasts of hypertrophic scar (HS), keloid and normal skin. Methods Fibroblasts of 5 generations in 6 case of HS, 6 keloid and 6 normal skin were inoculated in culture media containing IL-1 (200u/ml), 10 % serum DMEM, and cultured for 3 days. Avidin-biotin-peroxidase complex method (ABC) was used to show the expression of EGF, EGFR and TGFβ1. Results Density of EGF was 162.00±20.79 in fibroblasts of normal skin,151.00±26.45 in fibroblast of HS and 147.50±23.61 in fibroblasts of keloid. Density of EGFR was 148.34±13.72 in fibroblasts of normal skin, 136.67±17.52 in fibroblasts of HS and 131.67±11.71 in fibroblasts of keloid. Density of TGFβR 1 was 113.52±17.62 in fibroblasts of normal skin,106.71±21.43 in fibroblasts of HS and 107.54±23.52 in fibroblats of keloid. Density of EGF, EGFR and TGFβR 1 in fibroblasts of normal skin and keloid does not have significant difference (P>0.05). After IL-1β was addied, densities of EGF, EGFR, TGFβR 1 were 224.00±31.59, 178.67±27.86 and 80.34±11.75 respectively in fibroblasts of normal skin; 128.75±18.31, 105.82±21.61 and 109.83±25.79 in fibroblasts of HS;113.01±24.71,93.34±30.71 and 118.75±19.27 in fibroblasts of keloid. Densities of EGF and EGFR in fibroblasts of normal skin was significantly higher than those in HS and keloid (P<0.05). Densities of TGFβR 1 in fibroblasts of normal skin was significantly lower than those in HS and keloid. Conclusion IL-1β can increase the relative contents of EGF and EGFR, and decrease those of TGFβR 1 in fibroblast of the normal skin, and it can also down-regulate the expression of EGF and EGFR protein, but has no effect on densities of TGFβR 1 in fibroblasts of hypertrophic scar and keloid.
