川芎嗪对PC12细胞抗凋亡作用的实验研究

摘　　要：目的　探讨川芎嗪对多巴胺诱导的PC12细胞凋亡的保护作用及机制。方法　用多巴胺 (DA)刺激PC12细胞使其发生细胞凋亡 ,用流式细胞仪检测PC12细胞二倍体比率表征细胞凋亡率 ,用MTT法测定细胞活性 ,用Griess法间接测定NO的浓度 ,比较分析加入不同剂量的川芎嗪后对上述指标的影响。结果　加入 0 .3 0、0 .60mmol L的DA两组的PC12细胞的凋亡率、培养上清NO含量均显著高于无DA组 (P <0 .0 5 ) ,细胞活性显著低于无DA组 (P <0 .0 5 )。加入川芎嗪后 ,细胞凋亡率、NO含量均显著低于未加入川芎嗪组 (P <0 .0 5 )、细胞活性显著高于未加入川芎嗪组 (P <0 .0 5 )。结论　川芎嗪可抑制DA引起的PC12细胞凋亡 ,此作用可能与降低NO生成有关。Objective To explore the protective effect of ligustrazine on the apoptosis of PC12 cells induced by dopamine(DA) and its mechanisms. Methods Apoptosis of PC12 cell was induced by dopamine at the concentrations of 0, 0.30, 0.60 mM. Ligustrazine at the concentrations of 0, 30, 60, 90 μg/ml was added to defect the effect of ligustrazine on apotosis. The subdiploid peaks showing cell apoptosis rate were detected by flow cytometry. The cell activity was determined by MTT. Concentration of nitric oxide(NO) was determined by the Griess reaction. Results Compared with those of the control, the apoptosis rate and the level of NO induced by 0.30 or 0.60 mmol/L DA were higher, but the activity of PC12 cells was lower. Compared with those of the groups without ligustrazine, the apoptosis rate and the level of NO increased, but the cell activity decreased in a dose dependent manner. Conclusion Ligustrazine can inhibit the apoptosis induced by DA, which might be correlated with the decrease of NO production.
