摘 要:目的 :探讨新生鼠缺氧缺血再灌注后 ,c AMP反应元件结合蛋白 (CREB)磷酸化在海马的表达变化及其与海马神经元损伤后修复、存活的关系 .方法 :选用 7d龄SD鼠 16只 (不同窝 ) ,随机分入假术对照组、缺氧缺血脑损伤组 .经弹性管穿线法阻断右颈总动脉 3h ,予低氧 1h ;制备缺氧缺血脑损伤 (HIBD)模型 .3h后剪开扎线予再灌注 2 4h ,彩色多普勒监测血流 .应用免疫组织化学检测磷酸化的CREB (p CREB)在新生仔鼠HIBD再灌注 2 4h后海马区的表达 .结果 :HIBD组再灌注 2 4h仔鼠右侧海马CA1,CA3,DG区p CREB的表达明显增加 ,左侧相应区的增加低于右侧 ,但明显高于假术对照(P <0 .0 1) .缺血敏感区CA1低于耐受区CA3,DG(P <0 .0 1) .对照组p CREB两侧有基础表达 ,但无差别 .结论 :磷酸化CREB在HIBD再灌注 2 4h后海马区高表达 ,可能对保护CA1,CA3。AIM: To detect the expression of phosphorylation of c AMP response element binding protein (CREB) in the hippocampal neurons and to explore the relationship between the changes of p CREB and the survival and repair of neurons after the damage of hypoxic ischemia and reperfusion of the brain. METHODS: Seven day old SD rats ( n =12) which came from different broods were used in this study. Rats were randomly divided into sham group ( n =3) used as control group, hypoxic ischemia brain damage (HIBD) and 24hs’ reperfusion group ( n =9). A model of mild HIBD was established as follows: the right common carotid artery was temporarily occluded for 3 h through an elastic tube circling a thread. HIBD group was exposed to 80 mL·L -1 oxygen and 920 mL·L -1 nitrogen gas mixture for 1 h. After 3 h the tube was removed and the common carotid artery was reperfused. The flowing of blood was detected by the multicolor Doppler. At the 24 h reperfused timepoint, rats were perfusion fixed. The p CREB was immunohistochemically evaluated at different region of rats’ hippocampus. RESULTS: The CA1, CA3 and DG regions of right hippocampus in the HIBD group showed obvious immunoreactivity with the specific antibodies against p CREB ( P < 0.01 vs contral group). In the contralateral hippocampus of HIBD group, the immunopositive cells also increased, but less than that of the right side, but markedly higher than that of the control group. The expression of p CREB in CA1 region which was sensitive to hypoxic ischemia was slightly less than the resistant regions of CA3 and DG. The control group showed the basic immunoreactivity with the specific antibodies in the same regions. CONCLUSION: The high expression of p CREB in the hippocampus 24 h after hypoxic ischemia reperfusion may be very important in the protection of hippocampal pyramidal neurons in CA1, CA3 and DG.
