细胞因子对关节软骨细胞作用的研究

摘　　要：目的　观察白细胞介素 1β (IL - 1β)、肿瘤坏死因子α (TNF -α)、转化生长因子 β1(TGF - β1)对关节软骨细胞的影响 ,探讨骨性关节炎的发病机理。方法　应用关节软骨细胞体外培养 ,分别加入细胞因子 ,应用酶联免疫法 (ELISA)测定基质金属蛋白酶 - 3(MMP - 3)和前列腺素E2 (PGE2 )水平。结果　MMP - 3测定结果 ,细胞因子作用于软骨细胞 0～ 2 4h ,1、 10、 2 0ng/mlIL - 1β组 ,10ng/mlTNF -α组 ,高于对照组 (P <0 0 1) ;作用 2 4～ 4 8h ,1ng/mlIL - 1β组 ,0 1、 1、 10ng/mlTNF -α组 ,0 1、 1、 10ng/mlTGF - β1组低于对照组 (P <0 0 1)。 10ng/mlIL - 1β和 1ng/mlTGF - β1组低于10ng/mlIL - 1β组 (P <0 0 1)。PGE2 测定结果 ,细胞因子作用于软骨细胞 0～ 2 4h ,1、 10ng/mlTNF -α组高于对照组 (P<0 0 1) ,1ng/mlTNF -α和 1ng/mlTGF - β1组低于 1ng/mlTNF -α组 (P <0 0 1) ;作用 2 4～ 4 8h ,10、 2 0ng/mlIL - 1β组 ,0 1、 1、 10ng/mlTNF -α组高于对照组 (P <0 0 1)。 10ng/mlIL - 1β和 1ng/mlTGF - β1组低于 10ng/mlIL - 1β组 (P<0 0 1) ,1ng/mlTNF -α和 1ng/mlTGF - β1组低于 1ng/mlTGF -α组 (P <0 0 1)。 结论　细胞因子对软骨细胞分泌产生MMP - 3、PGE2 有较?Objective To explore the pathogenesis of arthritis,the effects of IL-1β,TNF-α,TGF-β 1 on joint chondrocyte were studied.Methods Joint chondrocyte was cultured in vitro, cytokines were added into the culture media respectively.The MMP-3 and PGE 2 levels in culture media were measured by use of ELISA.Results For MMP-3 level,after treated 0～24 hours,IL-1β at 1,10,20 ng/ml and TNF-α at 10 ng/ml were higher than that in control(P<0 01).After treated 24～48 hours,IL-1β at 1 ng/ml ,TNF-α at 0 1,1, 10 ng/ml and TGF-β 1 at 0 1,1,10 ng/ml were lower than that in control.IL-1β at 10 ng/ml & TGF-β 1 at 1 ng/ml was lower than that in IL-1β at 10ng/ml alone(P<0 01).For PGE 2 level,after treated with 0～24 hours,TNF-α at 1,10 ng/ml was higher than that in control(P<0 01),TNF-α at 1 ng/ml & TGF-β 1 at 1ng/ml was lower than that in at TNF-α 1ng/ml alone(P<0 01).After treated with 24～48 hours,IL-1β at 10,20 ng/ml,TNF-α at 0 1,1,10 ng/ml were higher than that in control(P<0 01).IL-1β at 10 ng/ml & TGF-β 1 at 1 ng/ml was lower than that in IL-1β AT 10ng/ml alone(P<0 01);TNF-α at 1 ng/ml & TGF-β 1 at 1ng/ml was lower than that in TNF-α at 1 ng/ml alone(P<0 01).Conclusion Cytokines could stimulate the expression of MMP-3 and PGE 2 significantly;TGF-β 1 could antagonize the effects of IL-1β and TNF-α apparently.
