鸡白细胞介素18成熟蛋白全长基因的克隆与序列测定

摘　　要：根据已发表的鸡白细胞介素 18(IL- 18)成熟蛋白的 c DNA序列 ,设计 1对特异性引物 ,应用 RT- PCR从脂多糖(L PS)刺激 10 h活化的 MDCC- MSB1细胞中克隆到编码鸡 IL - 18成熟活性蛋白的完整基因片段 ,其大小为 5 10 bp。经序列测定证实 ,所获得的 c DNA基因序列与文献报道的结果完全一致 ,从而为进一步研究鸡 IL- 18的基因表达、生物学活性以及应用奠定了基础。One pair of specific primer for mature chicken interleukin-18(ChIL-18) cDNA was designed and synthesized according to the previously published gene sequence of ChIL-18.The full length cDNA gene of ChIL-18 encoding mature active protein was amplified from LPS-stimulated MDCC-MSB1 cells by reverse transcription-polymerase chain reaction(RT-PCR).Then it was cloned into pMD18-T vector.Nucleotide sequence analysis of the positive recombinant plasmid showed that the obtained gene was a DNA fragment of 510 bp and the sequence of the cDNA was completely consistent with the reported ChIL-18 cDNA.This study provides the way for further study on the expression and biological function of ChIL-18.
