摘  要:本文初步建立了一种基于电泳分离和同步辐射X荧光(SRXRF)分析技术的生物样品内微量元素的种态分析方法。用同步辐射X荧光法直接分析了人肝细胞胞质溶胶等电聚焦(IEF)分离后蛋白条带内的金属锌含量。结果表明:肝细胞胞质溶胶内主要有三个含锌条带,等电点(pI)分别为4.69、5.93、9.84。含锌量分别占总检出锌的33.08%、17.05%、26.54%。其中等电点为4.69的蛋白可能为金属硫蛋白。另外在等电点为6.60—9.30的范围内有若干含锌蛋白,共占总检出锌的23.33%。A procedure based on isoelectric focusing ( IEF ) separation and synchrotron radiation X-ray fluorescence ( SRXRF ) analysis for speciation of trace elements in biological samples was preliminarily established. The proteins in human liver cytosol were separated by IEF, and zinc amount in each protein band was measured by SRXRF. The results indicate that there were three main zinc-containing bands with isoelectric points ( pIs) of 4.69, 5.93, and 9.84 present in the sample, and the zinc percentages in the three bands to the total recovered zinc were 33.08%, 17.05%, and 26.54%, respectively. In addition, several zinc-containing proteins were present in the pI range of 6.6 ? 9.3, which amounted to 23.33 % of the recovered zinc.
