摘  要:目的:探讨巢蛋白(nestin)在胚胎后期、新生期、成年3阶段的mRNA、蛋白水平表达的变化。方法:应用RT-PCR、免疫组化的方法进行半定量、定位分析。结果:在胚胎后期、新生期、成年3阶段Nestin在mRNA、蛋白水平均有表达,Nestin阳性细胞主要分布在胰岛中,腺泡周围的外分泌组织中有少量分布。结论:Nestin随大鼠胰腺的生长发育,核酸、蛋白表达下调,胰岛内Nestin阳性细胞百分数表达也下调。Objective:To study the expression of nestin and its mRNA in pancre as of different phases, including the embryonic, newborn and adult rats. Methods: The mRNA of nestin was detected by RT-PCR,the number of nestin-positive cells and the levels of nestin protein were accessed by the methods of immunohistoche mistry, and the results were analyzed by the statistical methods. Results:Nestin was expressed in E18.5,newborn and adult phases of rats. And nestin positive ce lls were mainly located in the islets and sparsely in the periacinar. Conclusion : The level of mRNA and protein of nestin in E18.5 was higher than that in new b orn and adult phase. The positive cells in islets in E18.5 was higher than adult phase. The nestin positive islet-derived cell in E18.5 may be a better resourc e for the research of stem cell than adult rat. [
