摘 要:目的 研究蛇毒神经生长因子 (Snakenervegrowthfactor,sNGF)对大鼠坐骨神经损伤修复的量效作用。方法 建立大鼠坐骨神经钳夹模型 ,术后肌注不同剂量 ( 3 0 0、90 0、2 70 0U kg)的sNGF ,通过展爪反射 ,趾间距 ,脊髓诱发电位、运动诱发电位观察 ,评定蛇毒神经生长因子对大鼠坐骨神经损伤修复的量效作用。结果 不同剂量sNGF明显促进受损周围神经运动和感觉功能的恢复 ,sNGF的上述作用与NGF的剂量有关。中剂量 90 0U kg效果较明显。Objective To investigate the dose effect of snake nerve growth factor(sNGF) on functional recovery of crushed sciatic nerves in rats. Methods Animal models of the crushed sciatic nerve were established in 50 Wistar rats and then divided into 5 groups: normal saline (NS) group, the low dose sNGF group, the moderate dose sNGF group, the high dose sNGF group and normal control group. sNGF was injected intramuscularly once a day for 14 d after crushing. Function repair for the crushed sciatic nerves was studied by the examination of somatosensory evoked potential(SEP), motor evoked potential(MEP) and the distance between the first and fifth toes(TS). Results At 2 weeks after crushing, SEP was enhanced by 26%, 50%, 86%, 90% in NS group, low dose group, moderate dose group and high dose group, respectively. There was significant difference between NS group and treated group ( P <0.01). There was also significant difference between low dose group and moderate dose group. However, there was no significant difference between moderate dose group and high dose group. Changes of MEP were the same as those of SEP. The TS was longer than that of the control group at 2 and 3 weeks in treated group. Significant difference was found between low dose group and moderate dose group and high dose group. Conclusion sNGF may repair the crushed nerve in rats in a dose dependent manner.
