摘 要:目的 :探讨早、晚静滴甲氨蝶呤 (MTX)时 ,其血药浓度及药动学参数的时辰变化 ,为制定我国患者的MTX给药方案提供依据。方法 :应用高效液相色谱法 ,测定 6例肿瘤患者接受早晚两个疗程化疗后的不同时间的血药浓度 ,并用 3P97软件进行模拟。结果 :静滴MTX后 ,其血药浓度存在明显的节律性 ,早上给药组药时曲线下面积AUC高于晚上给药组 ,其余药动学参数变化不明显。结论 :上午 8∶0 0~ 12∶0 0用药具有较好的效果。OBJECTIVE To study the difference of plasma drug level and pharmacodynamics while methotrexate (MTX) was administered during day and night. METHODS The concentrations of MTX in the serums of 6 patients were monitored at the different time using HPLC method. All data were simulated through 3P97 software. RESULTS The plasma concentration varied in a circadian rhythmic manner. Area under concentration curve (AUC) of MTX at day was significantly higher than that during might, No difference were observed in the other pharmacokinetics data. CONCLUSION It is better to use the MTX during the daytime.
