摘  要:合成了新荧光试剂 5 (4 羧基苯偶氮 ) 8 水杨醛缩氨基喹啉 (p CPSAQ ) ,探讨了与人血清蛋白反应的最佳实验条件。在 pH =7.0的缓冲体系中 ,蛋白质的存在使 p CPSAQ的荧光大大增强 ,据此建立了测定人血清蛋白质的灵敏方法 ;对人血清白蛋白和γ 球蛋白的线性范围分别为 0 .1~ 4 .5mg/L和 0 .1~ 4 .0mg/L ;检出限分别为0 .0 2 8和 0 .0 2 7mg/L。方法灵敏、快速、选择性好 ,用于人血清样品中总蛋白的测定 ,结果满意。New fluorescence reagent, 5-(4-carboxyphenylazo)-8-(4-methoxy)-benzylideneaminoquinoline (p-CPSAQ), has been synthesized, and was found to be a good fluorescence reagent for the determination of proteins. In neutral medium, p-CPSAQ was used as fluorescence probe in the determination of proteins, which was proved to be a simple, rapid and specific method. Linear relationship was found between the intensity of fluorescence and the concentration of protein in the range of 0.1~4.5 mg/L for human serum albumin (HSA) and 0.1~4.0 mg/L for γ-globulin (γ-IgG). The limits of detection were 0.028 mg/L for HSA and 0.027 mg/L for γ-IgG, respectively. The method has been applied to the analysis of human serum samples collected from the hospital and the results were in good agreement with those reported by the hospital. The method presented here is not only sensitive and simple, but also reliable and suitable for practical application.
